摘要
目的:研究黄芪总黄酮对自发性高血压大鼠肾脏的影响以及其可能的机制。方法:原发性高血压大鼠20只,随机均分为模型组和黄芪总黄酮治疗组,10只正常SD大鼠为正常对照组。取正常大鼠、原发性高血压大鼠及黄芪总黄酮治疗后的原发性高血压大鼠的肾脏,进行免疫组织化学染色和蛋白印迹实验,观察黄芪总黄酮对高血压大鼠TGF-β1、Smad2/3、Smad7的影响。结果:模型组与正常组比较,大鼠肾脏免疫组织化学染色及蛋白印迹显示TGF-β1、Smad2/3的表达显著升高(P<0.01),Smad7的表达显著降低(P<0.01);黄芪总黄酮治疗组与模型组比较,TGF-β1、Smad2/3的表达显著降低(P<0.01),Smad7的表达显著升高(P<0.01)。结论:黄芪总黄酮对高血压大鼠肾脏TGF-β1/Smad信号通路有显著影响,黄芪总黄酮可能通过调节TGF-β1/Smad信号通路产生了对高血压大鼠肾脏的保护作用。
To observe the impacts of total flavonoids of (TFA) on kidneys ofspontaneously hypertensive rats (SHR) and their possible mechanism. Methods: SHR were randomly divided into the model and TFA groups,the normal group was of 10 normal arterial pressure SD rats. Immunohisto- chemical staining and western blots were proceeded to determine kidney protein expression of TGF-^1, Smad2/3 and Smad7 in every group. Results: Compared with the normal group, the protein expression of TGF-^1 and Smad2/3 of the model group were significantly increased (P<0.01), and the expression of Smad7 were significantly reduced (P<0.01). After treatment with TFA, the protein expression of TGF-^1 and Smad2/3 in SHR were significantly reduced (P<0.01), and the expression of Smad7 were significantly increased (P<0.01). Conclusion: The TFA has remarkable influences on TGF-^1/Smad pathway in SHR kidney, it may produce protective effect on SHR kidney by adjust the TGF-p1/Smad signalings.
