摘要
目的:应用DNA扩增技术检验基础性材料,以构建DNA生物检验方法。方法:采用擦拭、剪取等方法对接触性材料进行取样,同一种材料采用三种方法进行检测,分别为DNA扩增法、QIAGEN硅膜提取法、Chelex-100提取法,比较三种方法检测结果的有效性。结果:DAN扩增技术在不同材料中的检出基因型均为单一来源,分别对5种材料多处去电进行扩增,DNA扩增法对各种接触性材料的成功率均高于QIAGEN硅膜提取法、Chelex-100提取法,QIAGEN硅膜提取法的成功率又高于Chelex-100提取法,组间差异具有统计学意义(P<0.05)。结论:DNA扩增技术对接触性材料具有较高的敏感性,是接触性材料检验的可靠方法。
Objective: To develop a method for the detection of DNA by DNA amplification. Methods: The contact material was sampled by wiping, clipping and so on. The same material was detected by three methods: DNA amplification method, QIAGEN silicon membrane extraction method and Chelex-100 extraction method. Results: The genotype of DAN in different materials was single source,and the DNA extraction method was more successful than QIAGEN in the contact of various materials. The success rate of the extraction method of silicon membrane, Chelex-100 and QIAGEN was higher than that of Chelex-100, and there was significant difference between groups (P<0.05). Conclusion: DNA amplification technology is highly sensitive to contact materials, and a reliable method for the inspection of contact materials.
