摘要
目的:探讨胆宁片对高脂饮食诱导的非酒精性脂肪肝大鼠SOD、MDA的影响。方法:SD大鼠53只,首先随机分为两组,正常组10只和模型组43只。通过高脂饮食诱导的方式复制非酒精性脂肪肝大鼠模型。造模第2周起每隔1周随机处死模型组中1只大鼠观察造模情况,于4周后确定造模成功,处死正常组10只,剩余40只模型组大鼠随机分为4组:模型组、胆宁片组、绞股蓝组、多烯磷脂酰胆碱组。各组分别予以相应剂量的药物灌胃治疗。连续给药4周后处死大鼠,生化法检测血清甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL),比色法检测肝组织匀浆中丙二醛(MDA)、超氧化物歧化酶(SOD)的含量。结果:治疗结束后,与模型组比较,胆宁片组能有效改善大鼠肝脏病理学状态(P<0.05);与模型组比较,胆宁片组大鼠血清中的血脂有不同程度的下降(P<0.01,P<0.05);胆宁片组肝组织MDA含量下降(P<0.01)、SOD含量升高(P<0.01),其中在升高SOD方面优于多烯磷脂酰胆碱组及绞股蓝组(P<0.01)。结论:胆宁片对改善高脂饮食诱导的NAFLD大鼠具有保护作用。
Objective:To explore the effect of Dan Ning tablet on SOD and MDA in rats with nonalcoholic fatty liver induced by high fat diet.Methods:53 SD rats were randomly divided into two groups:the normal group with 10 rats and the model group with 43 rats.The nonalcoholic fatty liver rat model was induced by high fat diet.From second weeks after modeling,one rat in the model group was randomly killed to observe the modeling situation for every other 1 weeks.The model was successfully established after4 weeks,and 10 rats in the normal group were killed.The remaining 40 rats in the model group were randomly divided into 4 groups:the model group,the Dan Ning tablet group,the gynostemma group,the polyene phosphatidylcholine group.The each groupswere given corresponding doses of drug gavage treatment.After 4 weeks of continuous administration,the rats were killed,and theserum triglyceride(TG),total cholesterol(TC),high density lipoprotein(HDL),low density lipoprotein(LDL) were detected by biochemistrymethod.The malonaldehyde(MDA),superoxide dismutase(SOD) content in the liver tissue homogenate were detected by colorimetry.Results:After the end of treatment,compared with the model group,the Dan Ning tablet group could effectively improveliver pathological state of rats(P<0.05).Compared with the model group,the serum lipid in rat serum of the Dan Ning tablet group had different degrees of decline(P<0.01,P<0.05).The liver tissue MDA content of the Dan Ning tablet group was decreased(P<0.01),and the SOD content was increased(P<0.01),in which the increase SOD was better than that of the polyenephosphatidylcholine group and the gynostemma group(P<0.01).Conclusion:Dan Ning tablets have protective effects on the improvement of NAFLD rats induced by high fat diet.
