低剂量ALA-PDT对人角质形成细胞生长和分泌细胞因子的作用研究 被引量:1

摘要 目的:研究不同剂量5-氨基酮戊酸-光动力治疗(5-aminolevulinic acid photodynamic,ALA-PDT)对体外培养的人角质形成细胞增殖的影响和分泌生长因子的作用。方法:首先以噻唑蓝(methylthiazolyldiphenyl-tetrazolium bromide,MTT)检测ALA-PDT对永生化人角质形成细胞(HaCaT细胞系)增殖的影响,再用相同的方法检测ALA-PDT对分离的人角质形成细胞(kerotinocyte,KC)增殖的影响,最后以ELISA法检测ALA-PDT对KC培养上清液中碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)和血管内皮生长因子(vascular endothelial growth factor,VEGF)含量的影响。结果:低剂量ALA-PDT(0.1m M和0.01m MALA,红光21J/cm^2)能促进HaCaT细胞和人角质形成细胞的增殖;高剂量ALAPDT(0.5m MALA,红光21J/cm^2)抑制其增殖。低剂量ALA-PDT干预24h后,bFGF分泌明显高于0.5m MALA组和单纯红光组;而VEGF呈现24h时轻度下降,24h后明显增加的趋势。结论:高剂量ALA-PDT抑制角质形成细胞的增殖,甚至杀伤细胞;低剂量ALA-PDT能促进角质形成细胞的增殖,并且促进角质形成细胞分泌bFGF和VEGF。 Objective To study effects of different doses of 5-aminolevulinic acid photodynamic (ALA-PDT) on the proliferation and secretion of growth factors in cultured keratinocytes. Methods The proliferation of HaCaT cell line and cultured human keratinocytes were detected by MTT. The contents of VEGF and bFGF in supernatant of cultured human keratinocytes were detected by ELISA. Results Low-dose PDT-ALA (0.1mM and 0.01mM) and 21J/cm2 red light-promoted the proliferation of HaCaT cells and human keratinocytes in vitro. High-dose ALA-PDT-0.5mM ALA and 21J/cm2 red light-inhibited the proliferation of HaCaT cells and human keratinocytes in vitro. After 24h treated by low-dose ALA-PDT, the secretion of bFGF in human keratinocytes was significantly higher than that in 0.5mM ALA group and red light group. The secretion of VEGF in human keratinocytes treated by ALA-PDT showed little change with in 24h. But it was obviously increased after24h. Conclusion High-dose ALA-PDT inhibited the proliferation of keratinocytes. Low-dose ALA-PDT could promote the proliferation of keratinocytes, and improve the secretion of bFGF and VEGF.
