大黄素对大鼠肾纤维化肾间质中骨形态发生蛋白-7表达的影响及意义

The effect and significance of emodin on expression of tissue inhibitor of bone morphogenetic protein-7 in rat with renal tubulointer-stitial fibrosis

目的:探讨大黄素对大鼠肾间质纤维化模型中骨形态发生蛋白-7(BMP-7)表达的影响及意义。方法:清洁级Wister雄性成年大鼠160只随机分为四组:正常对照组、假手术组、模型组及大黄素治疗组。制备单侧输尿管梗阻(UUO)模型[1];治疗组在UUO的基础上每天以大黄素40 mg/kg灌胃,分别在术后1 d、3 d、7 d、14 d,每组取10只大鼠处死,取左侧肾脏行免疫组织化学方法检测肾组织中BMP-7的表达水平,动态观察BMP-7在各组大鼠术后梗阻侧肾脏1 d、3 d、7 d、14 d的组织学变化及大黄素对其影响。结果:模型组BMP-7的表达持续减少;治疗组BMP-7的表达水平高于模型组(P<0.01)。结论:中药大黄素可以通过促进BMP-7的表达来发挥其延缓肾间质纤维化进程的作用,对肾纤维化有治疗作用。

Objective:To investigate the effect and significance of emodin on expression of tissue inhibitor of bone morphogenetic protein-7(BMP-7)in rat with renal tubulointer-stitial fibrosis.Methods:A total of160healthy Wister male rats were randomly divided into4groups:the normal control group,the sham-operation group,the UUO group and the emodin treated group(DTG).Rat in the DTG were administered intra-gastrically with emodin40mg/kg daily after modeling.Ten rats of each group were sacrificed respectively1,3,7and14days after UUO.Renal histopathological changes were observed by light microscope.The expression of BMP-7was examined by immunohistochemistry at each time point.Results:In UUO group,the expression of BMP-7in renal interstitium was decreased gradually.In DTG group,the expression of BMP-7increased significantly compared to that of the UUO group(P<0.01).Conclusions:Emodin could delay the progression of renal tubulointerstitial fibrosis and protect the function of renal tubule probably by up-regulating the expression of BMP-7.
