秋水仙素对洋紫荆种子萌发及幼苗生长的影响 被引量:6

Effects of Colchicine Treatment on the Seed Germination and Seedling Growth of Bauhinia variegate

摘要 以洋紫荆(Bauhinia variegata)成熟种子为试验材料,采用质量浓度为0.5、1.0、1.5、2.0 mg/m L的秋水仙素溶液分别处理12、24、36和48 h,并以蒸馏水浸泡种子作为对照,观察测定其种子萌发特性及幼苗生长指标。结果表明,随着秋水仙素质量浓度的增加,种子发芽率逐渐降低,2.0 mg/m L溶液使大部分种子致死,0.5、1.0 mg/m L质量浓度秋水仙素处理24 h有利于种子萌发;种子胚根的伸长随着秋水仙素质量浓度的增加也受到不同程度地抑制;秋水仙素对发芽种子胚芽的发育有一定的抑制作用;不同质量浓度秋水仙素处理后第7个月,各处理间苗高与地径差异均极显著(P<0.01),质量浓度为1.0 mg/m L,处理36h时,苗高和地径都最大;当秋水仙素质量浓度为2.0 mg/m L时,对叶片平均分裂角度有一定促进作用。 The mature seeds of Bauhinia variegate were studied by treating with0.5,1.0,1.5and2.0mg/mL colchicines solution for12,24,36and48h respectively,and the seeds soaked with distilled water were takenas control.The germination rate and the mutant seedling growth were observed after the treatments.The results showed that with the increase of colchicines concentration,seed germination rate decreased.2.0mg/mLcolchicines solution caused death of the seeds,and0.5,1.0mg/mL colchicines solution treatment for24h was themost beneficial to the germination of the seeds.With the increase of colchicines solution concentration,the radicleelongation was inhibited in different degrees;colchicine could cause the inhibition effect on continue developmentof germinating seed embryo.The change of seedling ground diameter was consistent with that of the seedlingheight after7months later,it showed that after soaking the seeds for36h in colchicine solution(1.0mg/mL),theseedling ground diameter and height were both the highest.High concentration(2.0mg/mL)colchicine solutionon average leaf split angle played an important role in germination promoting.The stimulative effect on leaf angle was found when the concentration of colchicines solution was2.0mg/mL.
