摘要
目的:研究长链脂肪乳剂对布比卡因所致心肌细胞膜流动性改变的影响。方法:采用NBD-C6-HPC特异性荧光探针标记心肌细胞膜脂质,借助激光共聚焦显微镜结合光脱色荧光恢复技术检测布比卡因及长链脂肪乳剂对心肌细胞膜流动性的影响。结果:对照(Con)组和1%体积分数长链脂肪乳剂(LCT)组比较,细胞膜流动性差异无统计学意义(P>0.05)。布比卡因1 000μmol/L(Bu-1000组)细胞膜的荧光恢复率与Con组、LCT组、布比卡因100μmol/L(Bu-100)组比较显著降低(P<0.01);Bu-1000组细胞膜的扩散系数显著低于Con组(P<0.05)和LCT组(P<0.01)。布比卡因1 000μmol/L+1%体积分数长链脂肪乳剂(Bu-1000+LCT)组细胞膜荧光恢复率(P<0.05)及扩散系数(P<0.01)显著高于Bu-1000组。布比卡因100μmol/L+1%体积分数长链脂肪乳剂(Bu-100+LCT)组细胞膜扩散系数(P<0.01)显著高于Bu-100组。结论:长链脂肪乳剂能够逆转布比卡因导致的心肌细胞膜流动性降低。
Objective: To study the effects of long-chain lipid emulsion on the changes of membrane fluidity induced by bupivacaine in H9c2 cardiomyocyte. Methods: By the combination of confocal laser scanning microscope and fluorescence recovery after photobleaching, the effects of long-chain lipid emulsion and bupivacaine on the membrane fluidity in H9c2 cardiomyocyte, whose membrane phospholipid was labelled with NBDC6-HPC specific fluorescent probes, were determined. Results: Comparing with the Con group, the LCT group showed no significant difference in membrane fluidity. The Bu-1000 group showed a decreased recovery ratio when compared with the Con group (P<0.01), LCT group (P<0.01) and Bu-100 group (P<0.01), and a decreased diffusion coefficient when compared with the Con group (P<0.05) and the LCT group (P<0.01). Compared with Bu-1000 group, the Bu-1000+LCT group showed an increased recovery ratio (P<0.05) and an increased diffusion coefficient (P<0.01). Compared with Bu-100 group, the Bu-100+LCT group showed an increased diffusion coefficient (P<0.01). Conclusion: Long-chain lipid emulsion can reverse the decrease of membrane fluidity induced by bupivacaine in H9c2 cardiomyocyte.
