摘要
以龙牙百合鳞片为材料,对其组织培养中的关键技术进行了研究。结果表明:百合鳞片组织培养最适表面消毒方法为75%乙醇擦洗3min+0.1%氯化汞浸泡13 min;最佳诱导培养基配方为MS+2,4-D 2.0 mg/L+6-BA 0.2 mg/L;最佳扩繁培养基配方为MS+6-BA 0.5 mg/L+NAA 0.5mg/L;最佳生根培养基配方为1/2MS+NAA 0.10 mg/L。
Taking Longya lilium bulb scales as materials, the key technique of tissue culture were studied. The result showed that the best disinfection method is scrub it with ethanol of 75% then steeped in mercuric chloride of 0.1 percent. When culture the Longya lily,optimize culture medium in each stage, there come to a conclusion that the culture medium of inducing callus of bulb scales is MS + 2,4-D 2mg/L+ 6BA 0.2mg/L, the culture medium of bulb rapid propagation is MS + BA0.5mg/L + NAA 0.5mg/L, the culture medium of take roots is 1/2MS + NAA 0.1mg/L.
