腹腔镜下射频消融联合静脉化疗对PTEN通路和Wnt通路介导肝癌细胞生长的影响

Influence of laparoscopic radiofrequency ablation combined with intravenous chemotherapy on hepatoma cell growth mediated by PTEN and Wnt pathway

目的探讨腹腔镜下射频消融术(LRFA)联合静脉化疗对PTEN通路、Wnt通路介导肝癌细胞生长的影响。方法收集2013年5月-2016年6月该院收治的肝细胞癌患者90例,按照随机数表法分为观察组(n=45)及对照组(n=45)。观察组患者接受LRFA联合静脉化疗,对照组患者仅接受静脉化疗。术后1周取两组患者的肝脏病灶组织标本,采用逆转录-聚合酶链反应法(RT-PCR)检测其中PTEN信号通路、Wnt信号通路相关基因m RNA表达量;采用放射免疫法检测血清肿瘤标志物含量。结果治疗1周后,观察组患者的病灶组织PTEN通路相关基因PTEN mRNA表达量高于对照组患者,缺氧诱导因子-1(HIF-1)和血管内皮生长因子(VEGF)mRNA表达量低于对照组患者,差异有统计学意义(P<0.05);观察组患者的病灶组织Wnt通路相关基因β-链蛋白(β-catenin)、细胞周期蛋白D1(Cyclin D1)、c-myc和基质金属蛋白酶7(MMP-7)mRNA表达量低于对照组患者,差异有统计学意义(P<0.05);观察组患者的血清肿瘤标志物甲胎蛋白(AFP)、γ-谷胺酰转肽酶同工酶Ⅱ(GGT-Ⅱ)、胰岛素样生长因子-2(IGF-2)、糖类抗原19-9(CA19-9)含量低于对照组患者,差异有统计学意义(P<0.05)。结论 LRFA联合静脉化疗可通过PTEN通路、Wnt通路抑制肝癌细胞生长,具有积极的临床意义。

Objective To study the influence of laparoscopic radiofrequency ablation combined with intravenous chemotherapy on hepatoma cell growth mediated by PTEN and Wnt pathway.Methods90cases of hepatocellular carcinoma patients from May2013to June2016were chosen as study subjects,all patients were divided into observation group(n=45),control group(n=45)randomly.Patients in observation group received laparoscopic radiofrequency ablation combined with intravenous chemotherapy,while those in control group received intravenous chemotherapy only.One week after treatment,two groups patients’liver lesion tissue PTEN signaling pathway,Wnt signaling pathway related gene mRNA expression were detected by RT-PCR method;serum contents of tumor markers were tested by Radioimmunoassay.Results One week after treatment,liver lesion tissue PTEN mRNA expression was higher in observation group than that in control group,HIF-1,VEGF mRNA expression were lower in observation group than those in control group,the difference was statistically significant(P<0.05);liver lesion tissue Wnt pathway related genes mRNA expression such asβ-catenin,CyclinD1,c-myc,MMP-7were lower in observation group than those in control group,the difference was statistically significant(P<0.05);serum contents of tumor markers such as AFP,GGT-II,IGF-2,CA19-9were lower in observation group than those in control group,the difference was statistically significant(P<0.05).Conclusion Laparoscopic radiofrequency ablation combined with intravenous chemotherapy can inhibit the growth of HCC cells through the PTEN and Wnt pathway,which has positive clinical significance.
