摘要
采用单因素试验法研究了蒙氏假单胞菌LKX-1菌株的发酵培养基组成和发酵条件,并采用正交试验法对发酵条件进行了优化。结果表明:适宜的发酵培养基配方为:麦芽糖2.0%,酵母粉2.0%,KH_2PO_4 0.15%,MgSO_4·7H_2O 0.10%;最适发酵条件为:发酵温度30℃,接种量1.0%,初始pH值7.0,装液量100 m L/瓶(三角瓶容量250 m L),发酵时间24 h;在此条件下,发酵液中活菌数可达4.5×10^(14)CFU/m L。
The composition of fermentation medium and fermentation conditions were studied by using single-factor test,and the fermentation conditions were optimized by using orthogonal experiment.The results showed that the optimal fermentation medium were as follows:maltose2%,yeast powder2%,KH2PO40.15%,MgSO4·7H2O0.10%,and the optimal fermentation conditions were as follows:fermentation temperature30℃,inoculation1%,the initial pH7.0,the liquid volume100mL/bottle,fermentation time24h.Under these conditions,the viable count reached4.5×1014CFU/mL.
