卡巴克络中间体氯乙酰儿茶酚的合成工艺研究

摘要 卡巴克络是一种常用的止血药,对毛细血管损伤流血有极为有效的止血效果.此药物的使用历史悠久,但原本的生产工艺中氯乙酰儿茶酚的合成使用剧毒物质一氯乙酸和氧氯化磷为反应原料,存在潜在危害,且氧氯化磷为酸性腐蚀品,对生产装置的要求高.因此通过对中间体氯乙酰儿茶酚合成工艺的研究,改用氯乙酰氯取代剧毒物质一氯乙酸和氧氯化磷,减少了反应潜在危害.经多次实验结果分析得出,原料(邻苯二酚颐氯乙酰氯)配比(摩尔比)、反应温度和反应时间3种因素对合成反应影响较大.并经过大量实验,得出结论:当原料配比为1颐1,反应时间为4 h,反应温度40益为最佳合成工艺.最佳合成工艺条件稳定,产率较高. Carbazochrome is a kind of common hemostatic,having an extremely effective hemostatic effect onthe capillary injury bleeding.The use of the drug has a long history.But in the original synthesis process ofchloroacetic catechol,the poisonous chloroacetic acid and phosphorus oxychlorideare used as raw materials.Thereare potential hazards,and oxygen phosphorus chloride is a corrosive acid,needing the high requirement ofequipment.So in this paper,chloroacetic chloride is used to replace the poisonous phosphorus oxychloride andchloroacetic acid to obtain chloroacetic catechol intermediate,reducing the potential hazards of reactions.Accordingto test results analysis,the three factors,namely raw material ratio(catechol:chloroacetyl chloride),reactiontemperature and reaction time,have a greater influence on the synthesis reaction.After a large number ofexperiments,the optimal synthesis technology is concluded that the ratio of raw materials(catechol:chloroacetylchloride),reaction time and reaction temperature are1:1,4h and40℃,respectively.The optimum synthetictechnology is stable and highly efficient.
