罗勒大型变种——千层塔植株再生体系的建立

摘要 探索罗勒组培快速繁殖技术,筛选植株再生体系的最佳培养条件,建立罗勒的高效植株再生体系。方法以罗勒种子为研究对象,采用不同的培养基开展丛生芽的诱导、继代增殖和生根培养。结果采用酒精和升汞对罗勒种子进行消毒,可获得无菌的试管苗。种子萌发的适宜培养基为MS培养基,试管苗增殖的适宜培养基为MS+6-BA(0.5 mg/L)+NAA(0.05mg/L)。在此过程中,试管苗极易玻璃化,应严格控制激素浓度,增大培养基的硬度。生根的最佳培养基为1/2MS+6-BA(0.5 mg/L)+NAA(0.05 mg/L)+IBA(0.05mg/L)。结论建立了罗勒的植株再生体系,筛选出罗勒种子消毒、萌发、试管苗增殖和生根的最佳培养基。 To explore rapid tissue cultivation propagation technology of Ocimum basilicum L, the optimal cultivation conditions were screened and the effective regeneration system of Ocimum basilicum L was established. Method: Ocimum basilicum L. seed as experiment object, buds induction, subculture and rooting of shoots were carried out on different media. Result: the seeds of Ocimum basilicum L. were disinfected with alcohol and mercuric chloride, sterile test tube planets were obtained. The suitable medium for seed germination was MS, and suitable growth medium was MS + 6-BA(0.5mg/L) + NAA(0.05mg/L). The best rooting medium was 1/2MS + 6-BA(0.5 mg/L) + NAA(0.05mg/L) + IBA(0.05mg/L). In the process, the test tube plantlet could extremely easily get vitrification. So the hormone concentration should be strictly controlled and the hardness of the cultivation medium should be increased. Conclusion: plant regeneration system of Ocimum basilicum L. was established; suitable conditions for seed of Ocimum basilicum L. germination, proliferation and rooting of plants were screened.
