鰤鱼诺卡氏菌与绛红色诺卡氏菌的融合菌研究 被引量:1

摘要 鰤鱼诺卡氏菌(Nocardia seriolae)为鱼类诺卡氏菌病的主要病原,绛红色诺卡氏菌(N.purpurea)是分离自土壤中一种无毒诺卡氏菌。以鰤鱼诺卡氏菌和绛红色诺卡氏菌为亲本进行原生质体融合,获取融合菌。经生长特征观察、革兰染色、16SrRNA测序分析,对融合菌进行初步鉴定,并通过斑马鱼模型评估融合菌毒力检测和免疫保护效果。结果表明,融合菌的菌落形态与亲本菌株不同,为诺卡氏菌属革兰阳性菌,融合菌的毒力较鰤鱼诺卡氏菌野生株降低70%,免疫保护率约为50%。 Nocardia seriolae,the main pathogen of fish nocardiosis,and N.purpurea,a non-toxic Nocardia sp.isolated from soil,were selected as parent strains for protoplast fusion.The fusion strains were identified by colony morphology,Gram staining and16s rRNA sequencing.Then the virulence test and immune protective effects of the fusion strains were evaluated by the zebrafish model.The results showed that the fusant was Gram-positive nocardia bacteria,and its colony morphology was different from the parent strains.The virulence of fusant decreased70%compared with wild strains N.seriolae ZJ0503and the immune protection rate of fusant is about50%.
