摘要 目的:探讨CTNNB1沉默对卵巢癌细胞增殖的作用。方法:用人类卵巢癌SKOV3细胞于体外培养,随机分为空白对照组、阴性对照组和CTN实验组,然后转染入靶向CTNNB1的shRNA载体质粒。通过Westernblot及MMT的方法检测CTNNB1的蛋白表达及细胞的活力。结果:以CTNNB1为靶向的shRNA能下调CTNNB1蛋白质在细胞中的表达(P<0.05);转染CTNNB1的shRNA后,CTN组的细胞生长活性与增殖受到了明显的抑制(P<0.05)。结论:CTNNB1基因的表达下调可抑制卵巢癌SKOV3细胞的增殖。 Objective:To investigate the effect of CTNNB1silencing on the proliferation of ovarian cancer cells.Methods:Humanovarian cancer SKOV3cells were cultured in vitro,and then randomly divided into the blank control group,the negative controlgroup and the CTN experimental group,and then transfected into shRNA vector plasmid targeting CTNNB1.Westernblot and MMTwere used to detect the expression of CTNNB1and cell viability.Results:CTNNB1targeting shRNA can down regulate theexpression of CTNNB1protein in cells(P<0.05).After transfection with CTNNB1shRNA,the cell growth and proliferation of CTNgroup were significantly inhibited(P<0.05).Conclusion:Down regulation of CTNNB1gene can inhibit the proliferation of ovariancancer SKOV3cells.
