摘要
目的:观察脂肪组织来源干细胞(ADSCs)对光老化皮肤中黑色素含量的降低作用。方法:将6周龄裸鼠(18~22g)双侧耳背部皮肤每2d紫外线光照一次,每次30min。持续4周后,通过观察皮肤变化和Fontana-Masson染色,确立光老化模型。每只裸鼠随机选取一侧耳背部皮下注射ADSCs溶液,对侧给予等量的平衡盐溶液(HBSS)。通过Fontana-Masson染色观察皮肤中黑色素含量。结果:ADSCs注射组黑色素少于对照组,差异具有统计学意义(P<0.05)。结论:光老化可导致皮肤黑色素含量增加,而脂肪干细胞可以降低光老化皮肤中黑色素的含量。
Objective To observe the efficacy of adipose derived stem cells(ADSCs)to reduce skin melanin content ofphotoaging.Methods The back skin of bilateral ears from6-week old nude mice(18-22g)was exposed to the ultravioletradiation once every two days,30min each time.Four weeks later,the successful establishment of the photoaging modelwas determined by observing the skin changes and Fontana-Masson dyeing.The ADSCs was randomly injected into a sideback subcutaneous area of ears using the micro syringe,while the other side was injected with equivalent Hank's Balanced Salt Solution(HBSS)as controls.Fontana-Masson stain was performed to observe the number of melanin in the skin.Results The number of melanin in ADSCs group was smaller than that of the control group,the difference was statisticallysignificant(P<0.05).Conclusion Photoaging can lead to an increase in melanin content of skin.ADSCs can reduce themelanin content of photoaging skin.
