紫河车提取物对人胶质瘤细胞增殖凋亡的影响

The effects of placental immunoegulating polypeptid on proliferation and apoptosis of human glioblastoma cells

目的:观察紫河车提取物对人脑胶质瘤细胞增殖与凋亡的影响。方法:把正常培养传代后的U251胶质瘤细胞按随机分配的方法分为四组,A组仅加普通培养液,B、C、D组各加浓度为100mg/ml、200mg/ml、400mg/ml的紫河车提取物5ml;观察显微镜下Trypan Blue染色后各组脑胶质瘤细胞的形态特点,MTT法观察测算U251细胞增殖情况,流式细胞仪检测U251细胞凋亡率。结果:B、C、D组细胞增殖指数逐渐下降,与A组进行比较,各组P均<0.05;将各组培养24h后上机,测得A、B、C、D组细胞凋亡率分别为(0.2±0.2)%、(8.6±1.8)%、(17.9±3.3)%、(28.2±3.1)%。其中,B、C、D组和A组进行比较,P均<0.05。结论:紫河车提取物可抑制人脑胶质瘤细胞增殖,并诱导其凋亡。

Objective:To observe the effects of placental immunoegulating polypeptid on proliferation and apoptosis of humanglioblastoma cells.Methods:Normal U251glioma cells were randomly divided into four groups.Group A was only given nutrient solution.Group B,group C and group D were all given5ml placental immunoegulating polypeptid,100mg/ml,200mg/ml,400mg/ml respectively.The form of cells of trypan blue stained U251glioma cells was observed under the microscope.MTT assay was used to measure theproliferation of U251cells.The apoptotic rate of U251cells was detected by flow cytometry.Results:The index of cell proliferation ingroup B,group C and group D were declined gradually,P<0.05.Cultured for24h,cell apoptosis in group A,B,C and D were(0.2±0.2)%,(8.6±1.8)%,(17.9±3.3)%,(28.2±3.1)%respectively.Compared with group A,P<0.05.Conclusion:Placental immunoegulatingpolypeptid can restrain the proliferation of U251glioma,and also lead to the apoptosis of U251cells.
