克罗恩病患者蛋白质组学差异表达的研究

Proteomic differential expression in patients with Crohn's disease

目的探讨克罗恩病患者血清中的差异表达蛋白。方法选取在本院诊治的克罗恩病患者20例,采用各种Cy Dve染料实施交叉标记后,依据顺序实施MALDI-TOF-MS、2-D DIGE等鉴定操作。结果对二者双向电泳图对比,发现克罗恩病患者在表达异常蛋白质点方面有多个,其中1 058蛋白质点,相比于正常成人组,则存在明显升高,即1.67倍(P<0.05),此蛋白质通过质谱鉴定得知,乃是一种结合珠蛋白质。结论在克罗恩病血清中,结合珠蛋白所存在的高表达,提示在其病程中免疫系统失衡可能存在一定作用。

Objective To investigate the differences in serum of patients with Crohn disease protein.Methods20cases in our hospital for treatment of Crohn's disease patients,the implementation of cross labeled by various CyDve dyes,the implementation of MALDI-TOF-MS based on2-D DIGE sequence identification operation.Results Of the two dimensional electrophoresis map comparison,found that patients with Crohn's disease the abnormal expression of protein spots are multiple,including1058protein spots,compared to the normal adult group,there is increased1.67times(P<0.05),the protein identification by mass spectrometry that is a combination of pearl protein in serum.Conclusion High expression of Crohn's disease,combined with the existing pearl protein,suggesting that in the course of immune system imbalance may have some effect.
