山楂叶原花青素对肿瘤坏死因子-α诱导人脐静脉内皮细胞细胞间黏附分子-1和E-选择素过表达的影响 被引量:3

摘要 目的观察山楂叶原花青素对肿瘤坏死因子-α(TNF-α)诱导的人脐静脉内皮细胞细胞间黏附分子-1(ICAM-1)和E-选择素过表达的影响,探讨其抗炎作用的机制。方法体外培养人脐静脉内皮细胞。MTT法检测细胞活力,流式细胞术检测ICAM-1和E-选择素的表达。结果山楂叶原花青素浓度≤200 mg/L时,对细胞活力无明显影响;模型组人脐静脉内皮细胞ICAM-1和E-选择素表达较正常组显著升高,10、20、30、40、50 mg/L山楂叶原花青素浓度依赖地抑制ICAM-1和E-选择素的表达升高,其中40、50 mg/L组较模型组差异有统计学意义。结论山楂叶原花青素能够抑制TNF-α诱导的血管内皮细胞ICAM-1和E-选择素的过表达,这可能与其抗炎作用相关。 Objective To observe the effects of hawthorn leaf procyanidins(HLP)on over expressions of ICAM-1and E-selectin in human umbilical vein endothelial cells(HUVEC)induced by TNF-α,and clarify the mechanism of HLP's anti-inflammation effect.Methods HUVEC were cultured in vitro.MTT assay was used to detect cell viabilities.The expressions of ICAM-1and E-selectin in HUVEC were detected by flowcytometry.Results Up to200mg/L,HLP showed no significant decrease in cell viabilities;the expression levels of ICAM-1and E-selectin in the model group significantly increased,compared with that in the normal group;10,20,30,40,50mg/L HLP inhibited the expression elevations of ICAM-1and E-selectin in concentration-dependent manner;and there were statistical significances in40,50mg/L HLP groups,compared with the model group.Conclusion HLP can inhibit the over expressions of ICAM-1and E-selectin of vascular endothelial cells induced by TNF-α,which possibly underlies HLP's anti-inflammation effect.
