金丝桃素对抑郁症大鼠海马5-HT及5-HT1A受体表达的影响 被引量:4

摘要 目的本研究旨在探讨贯叶金丝桃有效成分之一的金丝桃素对抑郁症模型大鼠海马5-HT及5-HT1A受体表达的影响。方法将45只SD大鼠,分为对照组、模型组和给药组。采用慢性轻度不可预刺激(CUMS)抑郁大鼠模型,对照组和模型组给生理盐水,给药组给予金丝桃素。采用强迫游泳实验、悬尾实验进行行为学检测,采用高效液相色谱检测5-HT浓度,Westernblot实验检测5-HT1A受体的表达。结果金丝桃素能够降低强迫游泳实验及悬尾实验中的静止时间(P<0.05),升高了海马5-HT的含量,增强了5-HT1A受体的表达(P<0.05)。结论金丝桃素抗抑郁作用可能与海马5-HT水平及5-HT1A受体表达有关。 Objective:The purpose of this study was to investigate Hypericin,one of the active ingredient of Hypericum perforatum L.,on5-HT level and expression of5-HT1A receptor in hippocampus of depressive rats.Methods:45SD rats were randomly divided into control group,model group and treatment group.The depression animal model was produced by using unpredictable chronic mild stimulation(CUMS)model,and the control group and model group were given saline,the treatment group was administered with hypericin.The depressional behavior was examined using forced swimming test and tail suspension test.The level of5-HT and expressionof5-HT1A receptor protein were detected using high performance liquid chromatography(HPLC)and Western blot respectively.Results:The rest time can be reduced by hypericin in forced swimming test and tail suspension test(P<005),the content of5-HT in the hippocampus increased and the5-HT1A receptor expression enhanced(P<005).Conclusion:Antidepressant effect of hypericin may be related to the level of hippocampal5-HT and5-HT1A receptor expression.
