食醋干预下大鼠血清对食管癌EC109细胞增殖活性的影响

摘要 目的研究食醋干预下SD大鼠血清对食管癌EC109细胞增殖活性的影响。方法Ⅰ组(对照组):以含20%灭活普通SD大鼠血清血清培养液培养食管癌EC109细胞;Ⅱ组(实验组):以含20%灭活长期高醋饮食干预条件下SD大鼠血清培养食管癌EC109细胞;细胞生长曲线、细胞DNA合成以及细胞周期分布应用用MTT比色法和流式细胞术检测。计算各个时间点抑制率、细胞S期分数和增殖指数;结果2组间不同时间点OD值有显著差异性,有统计学意义(P<0.05)。2组不同时间点抑制率的统计分析结果显示差异有统计学意义(P<0.05)。Ⅰ组细胞G0/G1期细胞比例低于Ⅱ组,而S期及G2/M期细胞所占的比例明显高Ⅱ组;Ⅱ组S期分数及细胞增殖指数显著低于Ⅰ组,差异有统计学意义(P<0.05)。结论长期高醋饮食干预条件下SD大鼠血清能抑制食管癌EC109细胞的生长以及DNA合成。食醋对食管癌EC109细胞的增殖具有明显的抑制作用。 Objective:Study the effect of using the SD Rat′Serum to inhibit the esophagus cancer EC109cell proliferative activity under the vinegar intervention.Methods:Ⅰgroup(control group):Using the20%inactivation common SD Rat′Serum to cultivate esophagus cancer EC109cells;Ⅱgroup(experimental group):Using the20%inactivation SD Rat,Serum under the condition of long-term vinegar diet intervention to cultivate esophagus cancer EC109cells;MTT Colorimetric method and flow cytometry detection method were used to measure the cell growth curve,cell DNA synthesis and cell cycle distribution,then calculate the every point-in-time inhibition rate,S phase grade and cellular proliferation index.Results:There was a significant difference between the different time point OD value of two groups(P<0.05).Statistical analysis shows that the different point-in-time inhibition rate had the significant statistical difference(P<0.05).TheⅡgroup G0/G1phase cell proportion was much higher than theⅠgroup,but the S phase and G2/M phase cell proportion were much lower than theⅠgroup;The S phase grade and cellular proliferation index of theⅡgroup were much lower than theⅠgroup;The difference had the statistical significance(P<0.05).Conclusion:The SD Rat′Serum under the condition of long-term vinegar diet intervention can inhibit the grow and DNA synthesis of the esophagus cancer EC109cell.The vinegar has significient inhibitory effect to the esophagus cancer EC109cell.
