参草通脉颗粒对慢性心衰大鼠MMP-3与TIMP-3的影响的实验研究

Experimental study on effects of Shencao Tongmai granule on MMP-3 and TIMP-3 in chronic heart failure rats

目的探讨参草通脉颗粒对慢性心衰大鼠MMP-3与TIMP-3的影响。方法随机选取10只大鼠作为正常对照组,10只作为假手术组,40只构建慢性心衰模型,建模成功后随即选取30只,分为模型组、西药组及中药组,各10只;对照组、假手术组及模型组采用蒸馏水灌胃,西药组给予赖诺普利处理,中药组给予参草通脉颗粒处理;Western blot法检测心肌组织中MMP-3、TIMP-3蛋白表达量。结果与正常对照组相比,假手术组MMP-3表达有所增加,TIMP-3表达有所下降,但组间比较差异不显著(P>0.05);与假手术组相比,模型组MMP-3表达显著增加,TIMP-3表达显著下降(P<0.05);中药组及西药组与模型组相比,MMP-3表达显著下调且TIMP-3表达显著上调(P<0.05),但并未恢复到假手术组水平;中药组与西药组组间比较MMP-3及TIMP-3表达差异不显著(P>0.05)。此外,中药组、西药组与模型组相比,LVIDs、LVIDd下降,EF值上升(P<0.05)。结论参草通脉颗粒可以下调MMP-3的表达,增加TIMP-3的表达,从而阻止MMP-3对心肌细胞外基质结构的破坏作用,阻止CHF的发展。

Objective To explore the effects of Shencao Tongmai granule on MMP-3and TIMP3expressions in chronic heart failure rats.Methods10rats were randomly selected as control group,other10as sham-operated group,and40rats were chosen underwent chronic heart failure model building,within which30were chosen as model group(10cases),western medicine group(10cases),and traditional Chinese medicine group(10cases),randomly.The control group,sham-operated group and model group were given gastric perfusion of distilled water.Western medicine group was taken lisinopril and the traditional Chinese medicine group was taken Shencao Tongmai granule.The expressing quantities of MMP-3and TIMP3in myocardial tissue were detected by Western blot.Results Compared with control group,the expression of MMP-3in sham-operated group was increased and TIMP-3decreased(P>0.05).Compared with sham-operated group,the expression of MMP-3in model group was increased and TIMP-3decreased(P<0.05).Compared with western medicine group and traditional Chinese medicine group,the expression of MMP-3was decreased and TIMP-3increased(P<0.05).There was no significant difference of MMP-3and TIMP-3between western medicine group and traditional Chinese medicine group(P>0.05).In addition,compared with model group,LVIDs and LVIDd decreased and EF values increasedin the traditional Chinese medicine group and western medicine group(P<0.05).Conclusion Shencao Tongmai granule can reduce the expression of MMP-3and promote the expression of TIMP-3,so as to stop the destruction of MMP-3to myocardial extracellular matrix and to impede the progress of CHF.
