摘要
目的研究并探讨冰冻红细胞的制备方法及应用价值。方法对100份悬浮红细胞(2016年6月~2017年1月本站采集2~6d内的RH(D)阴性红细胞16U)标本在不同条件下进行制备,制备方法为甘油化、去甘油化,分别在不同放置时间、不同离心速度下将其制备成冰冻红细胞,比较加甘油后不同放置时间的甘油残留量、去甘油化过程中不同离心速度的各项冰冻红细胞参数。结果加甘油后不同放置时间下,放置30min的标本甘油残留量明显低于放置15min的标本(P<0.05);不同离心速度下,去甘油化后冰冻解冻红细胞的各项参数均符合国家质检标准,离心速度2600转/min的标本其甘油残留量、体外溶血率、游离血红蛋白均明显低于离心速度3300转/min的标本(P<0.05),且其红细胞回收率明显高于离心速度3300转/min的标本(P<0.05)。结论对红细胞标本进行甘油化、去甘油化制备,可获得优质冰冻红细胞,有利于保证输血的安全性,缓解血液供需矛盾。
Objective To study and explore the preparation method and application value of frozen red blood cells.Methods100suspension with white and red cell specimens(RH(D)negative erythrocyte2-6d(16U)were collected in this station from June2016to January2017)were prepared under different conditions.The frozen red blood cells were preparated by the preparation method of glycerol and glycerol,respectively in different time and different centrifugal speed under,at different times after the addition of glycerol for glycerol residues,red blood cell the parameters of different centrifugal speed in the process of freezing glycerol.Results The time of different placement with glycerol,glycerol residues placed specimens of30min were significantly lower than that of15min(P<0.05)were placed.Different centrifugal speeds,deglycerolizing after the parameters of frozen thawing red blood cell were in line with national quality inspection standards,centrifugal speed2600rpm/min were the glycerol residue and residual white blood cells in vitro,and free hemoglobin were significantly lower than that of centrifugal speed3300rpm/min samples(P<0.05),and the recovery rate of RBC was significantly higher than that of centrifugal speed3300rpm/min samples(P<0.05).Conclusion The red blood cells can be obtained by high quality frozen red blood cells.It is helpful to ensure the safety of blood transfusion and relieve the contradiction between supply and demand of blood.
