摘要
目的观察氟西汀对皮质酮诱导的SH-SY5Y细胞株中下丘脑摄食调节因子Nesfatin-1含量升高的影响。方法采用体外培养细胞的方法,建立SH-SY5Y细胞皮质酮损伤模型,设氟西汀给药组(10^(-9)~10^(-5)mol·L^(-1))、皮质酮组和阴性对照组,采用实时荧光定量PCR、Western blot和免疫组化的方法观察细胞中Nesfatin-1 mRNA及蛋白表达的变化。结果与对照组相比,皮质酮组细胞Nesfatin-1 mRNA和蛋白含量均显著升高;氟西汀(10^(-9)~10^(-5)mol·L^(-1))处理后Nesfatin-1 mRNA和蛋白含量均显著降低。结论氟西汀体外用药能够逆转皮质酮诱导的SH-SY5Y细胞Nesfatin-1表达上调。
Objective To investigate the effect of fluoxetine on the increased content of Nesfatin-1induced by corticosterone in the human neuroblastoma SH-SY5Y cell.Methods Using SH-SY5Y cell line to establish corticosterone-induced SH-SY5Y cell injury model.We designed five groups of fluoxetine(109-105mol?L!),corticosterone group and control group.Real-time PCR,western blot and immunohistochemistry were used to measure the mRNA and protein expressions of Nesfatin-1.Results The mRNA and protein expressions of Nesfatin-1in the corticosterone group were higher than those of the control group.The mRNA and protein expressions of Nesfatin-1in the fluoxetine group were lower than those of the corticosterone group.Conclusion The increased content of Nesfatin-1induced by the corticosterone treatment could be decrease by the fluoxetine treatment.
