摘要
目的探讨<3岁反复喘息患儿外周血血清类黏蛋白1样蛋白3(ORMDL3)基因在不同临床表型中的表达及其与血清白介素-17(IL-17)和嗜酸性细胞阳离子蛋白(ECP)的关系。方法选取该院儿科就诊<3岁反复哮喘的患儿87例,根据患儿是否存在高危因素分成特应性哮喘组(64例)和非特应性哮喘组(23例),再选取同期在儿科体检的健康儿童30例。分析3组研究对象ORMDL3基因表达水平、血清IL-17和ECP含量和ORMDL3基因频率分布的差异,并对3组研究对象ORMDL3基因表达水平和血清IL-17和ECP含量的相关性进行分析。结果特应性哮喘组ORMDL3基因表达水平(11.57±1.96)%,血清IL-17(1 354.46±873.48)pg/ml和血清ECP(45.65±12.76)μg/L均高于非特应性哮喘组和对照组(P=0.000);3组的ORMDL3基因AA、AG和GG 3种基因型的频率分布差异有统计学意义(χ~2=12.84,P=0.012)。特应性哮喘组的ORMDL3基因表达水平与血清IL-17(r=0.318,P=0.000)、血清ECP(r=0.540,P=0.000)均呈正相关。结论不同临床类型的哮喘患儿血清中的ORMDL3基因表达水平、IL-17和ECP含量不同。特应性哮喘患儿的血清ORMDL3基因表达水平与血清IL-17、ECP含量存在正相关性。
Objective To investigate the expression of orosomucoid1-like protein3(ORMDL3)gene of different clinical phenotypes and its relationship between seruminterleukin-17(IL-17),serum eosinophil cationic protein(ECP)in peripheral blood of the recurrent asthma patients no more than3years old.Methods Eighty recurrent asthma children no more than3years old admitted to our hospital were selected and divided into atopic asthma group(64cases)and non-atopic asthma group(23cases)according to the existence of high risk factors.In the same period30cases of healthy children were selected in the control group.The differences of ORMDL3gene expression,serum IL-17,serum ECP,and ORMDL3gene frequency distribution in the three groups were analyzed,and the correlations of ORMDL3gene expression level with serum IL-17content,serum ECP content in the3groups were analyzed.Results The expression levels of ORMDL3gene(1354.46±873.48)pg/ml and serum ECP content(45.65±12.76)ug/L in the atopic asthma group were higher than those in the non-atopic asthma group and the control group(P=0.000).The distribution of AA,AG and GG genotypes of ORMDL3had no significant difference among the3groups(x2=12.84,P=0.012).The expression levels of ORMDL3gene was positively related with serum IL-7(r=0.318,P=0.000)and serum Eop(r=0.540,P=0.000)in the atopic group.Conclusions The serum expression levels of ORMDL3gene,ECP and IL-17are different among asthma children with different clinical phenotypes.The expression level of ORMDL3gene in the serum of atopic asthma patients no more than3years old is related to the levels ofserum IL-17and ECP.
