摘要
目的:研究远志多糖对由东莨菪碱所致小鼠空间学习记忆障碍的影响及相关作用机制。方法:采用水提醇沉法得到远志多糖,利用东莨菪碱诱导小鼠学习记忆障碍模型共4组(正常组、模型组、阳性对照组、远志多糖组);采用ELISA试剂盒测定ACh和BDNF含量,综合评价远志多糖对小鼠学习记忆的改善效果。结果:利用东莨菪碱成功诱导小鼠学习记忆模型,在给予远志多糖组治疗后,ACh和BDNF含量显著增加,且高于阳性对照组。结论:远志多糖通过增加脑内ACh和BDNF的含量,能够显著改善小鼠学习记忆障碍,促进小鼠学习、记忆能力的提高,可能与胆碱酯酶抑制剂改善学习记忆的机制有关。
Objective:To study the effect of polygala tenuifolia polysaccharides on spatial learning and memory impairment induced by scopolamine in mice and its related mechanism.Methods:Water extract and alcohol precipitation were used to obtain polygala tenuifolia,using scopolamine to induce model of learning and memory impairment in mice.They were divided into4groups(the normal group,the model group,the positive control group,the polygala polysaccharide group).The determination of Ach and BDNF were detected by ELISA kit,and the comprehensive evaluation of polygala polysaccharide on mice learning and memory improvement was carried out.Results:We successfully induced learning and memory in mice model by scopolamine.After given the polygala polysaccharide group treatment,the ACh and BDNF were increased significantly,and higher than the positive control group of oxiracetam.Conclusion:Polygala polysaccharide by increasing brain levels of Ach and BDNF can significantly improve the learning and memory impairment in mice,promote the learning and memory of mice increased,which may improve the mechanism of learning and memory with different cholinesterase inhibitors.
