血清抵抗素水平及抵抗素基因启动子-420C/G多态性与川崎病的相关性研究 被引量:3

摘要 目的观察血清中抵抗素水平及抵抗素基因启动子-420C/G多态性在川崎病的表达水平,并对其相关性进行分析。方法随机选择60例川崎病(KD)患儿为KD组,另设60例健康儿童为对照组。两组均采用酶联免疫吸附实验检测儿童血清抵抗素水平,应用多聚酶链反应限制性片段长度多态性技术检测不同组别患儿抵抗素基因-420C/G位点多态性的差异,同时检测统计其血液学相关指标,包括血白细胞计数(WBC)、红细胞计数(RBC)、血红蛋白水平(Hb),红细胞压积(Hct)。结果与对照组比较,KD患儿外周血液中WBC、RBC、Hb及Hct表达水平均降低(P<0.05),而血小板计数(PLT)表达水平升高(P<0.05),差异有统计学意义;血清抵抗素检测结果显示,与对照组相比,KD患儿血清抵抗素水平升高;限制性核酸内切酶试剂盒BpiⅠ检测血清抵抗素基因启动子-420C/G多态性,与对照组相比,KD组患儿GG较CC基因频率较低(P<0.05)。Pearson相关分析显示,KD患儿血浆抵抗素水平与WBC、RBC、Hb及Hct呈负相关(P<0.05),而与PLT表达水平呈正相关(P<0.05);KD患儿组的GG基因型的WBC、RBC、PLT、Hct及血清抵抗素比CG基因型高,GG、CC基因型均高于CC基因型。结论在川崎病患儿血清抵抗素水平升高,提示血清抵抗素水平的检测可作为川崎病诊断的潜在可靠指标。 Objective To observe the serum resistin level and resistin gene promoter420C/Gpolymorphism in Kawasaki disease(KD),and to analyze its relevance.Methods Sixty children with Kawasakidisease(KD)were randomly selected for KD group,the same number of healthy children were included intocontrol group.ELISA was used to detect serum resistin level in the children.PCR restriction fragment lengthpolymorphism(PCR-RFLP)technique was utilized to detect the polymorphisms of resistin gene-420C/G locus.The related parameters of hematology including blood leucocyte count(WBC),red blood cell count(RBC),hemoglobin(Hb),and hemotocrit(Hct)were also tested.Results Compared with the control group,theperipheral blood levels of WBC,RBC,Hb and Hct were significantly decreased,and the platelet count wassignificantly increased in the KD group(P<0.05).The serum resistin level of the KD group was significantlyhigher than that of the control group.Detection of serum resistin gene promoter420C/G polymorphism showedthat compared with the control group,the gene frequency of GG was relatively lower than that of CC in theKD group(P<0.05).Pearson correlation analysis showed that in the children with KD,the plasma resistin level was in negative correlation with WBC,RBC,Hb and Hct(P<0.05);but in positive correlation withplatelet count(P<0.05).WBC,RBC,platelet,Hct and resistin levels in the KD children with GG genotypewere significantly higher than those in the KD children with CG genotype,which were in turn significantlyhigher than those in the KD children with CC genotype.Conclusions Serum resistin level significantlyincreases in children with Kawasaki disease,which suggests serum resistin level can be used as a potentialindicator for diagnosis of Kawasaki disease.
