摘要
目的建立超高效液相色谱(UPLC)方法 ,测定紫杉醇注射液中紫杉醇的含量。方法采用Hypersll BDS C18色谱柱(2.1 mm×100.0 mm,3μm),以甲醇-水(55∶45,V/V),分析波长230 nm,流速0.4 ml/min,柱温30℃,进样量10μl,等度洗脱。结果紫杉醇在0.5~50μg/ml范围内具有良好的线性关系(R2=0.9992),经过方法学验证,符合要求。结论本实验建立的UPLC方法可以简便、快速地可用于紫杉醇的含量测定。
Objective To determine the content of paclitaxel injection by establishment of ultra performance liquid chromatography(UPLC)method.Methods The wavelength was230nm,with flow rate of0.4ml/min,column temperature of30℃,injection amount of10L and isocratic elution,determined by Hypersll BDS C18chromatographic column(2.1mm×100.0mm,3μm)and methanol-water(55∶45,V/V).Results Paclitaxel had a good linear relationship in the range of0.5~50μg/ml(R2=0.9992),and it had been verified by the method and meets the requirements.Conclusion The UPLC method established in this experiment can be used for the determination of paclitaxel in a simple and rapid manner.
