摘要
[目的]研究二溴海因复合消毒剂(主要成分为二溴海因(DBDMH)和邻苯二甲醛(OPA))的消毒性能及杀菌机理,开发适合口岸使用的广谱高效消毒剂。[方法 ]用悬液定量杀菌实验,评估二溴海因复合消毒剂的杀菌效果;利用傅里叶红外光谱法和流式细胞法,研究消毒剂的杀菌机理。[结果 ]复合消毒剂对大肠杆菌、铜绿假单胞菌、金黄色葡萄球菌的作用浓度(杀灭对数值大于5)分别为:100 mg/L DBDMH+50 mg/L OPA、60 mg/L DBDMH+210 mg/L OPA、200 mg/L DBDMH+50 mg/L OPA。复合消毒剂能引起大肠杆菌外部形态变化,导致生物大分子上的羰基、亚氨基、磷酸二酯键和糖苷键不同程度断裂,使得ATP含量迅速下降,膜电位变化。[结论 ]复合消毒剂对细菌的杀灭效果较好。
[Objective]To investigate the disinfection performance and mechanism of Dibromohydantin combinationdisinfectant(Dibromo-5,5-dimethylhydantoin,DBDMH,Ortho-phthalaldehyde,OPA),and to develop a broadspectrum and high efficiency disinfectant suitable for port use.[Methods]Suspension quantitative sterilization testwas used to examine the germicidal efficacy of the combination Dibromohydantin disinfectant,and the bactericidalmechanism of E.coli was investigated after treating by the disinfectant through Fourier Transform Infrared Spectrometerand Flow cytometer.[Results]The average killing logarithm was above5when E.coli,p.aeruginosa,S.aureu wereexposed in100mg/L DBDMH+50mg/L OPA,60mg/L DBDMH+210mg/L OPA,200mg/L DBDMH+50mg/L OPAseparately.The external morphological of E.coli changed,which caused the carbonyl group,the imino group,thephosphate two ester bond and the glycosidic bond of the biological macromolecule were broken in different degrees.Thecontent of ATP decreased rapidly and the membrane potential changed.[Conclusions]Dibromohydantin combinationdisinfectant has better effect on bacteria.
