放射性直肠纤维化组织中lncRNA差异性表达研究

摘要 目的探讨长链非编码RNA(lncRNA)在放射性直肠纤维化(RIRF)组织中的表达变化。方法采用高通量基因芯片检测长程放疗后的直肠组织中lncRNA与未行放疗的正常直肠组织中的表达差异,分析lncRNA的表达变化与RIRF的关系。结果放疗后直肠纤维化组织较正常直肠组织中表达量相差2倍以上的lncRNAs共1622个,其中1038个上调,584个下调,表达量相差10倍以上lncRNAs共76个,其中54个上调,22个下调。生物信息学技术显示多个差异性表达的lncRNAs参与调节纤维化信号通路相关细胞因子,如FGF2、TGF-β1等。结论 lncRNA可能通过调控纤维化信号通路的细胞因子参与RIRF的发生,有望成为RIRF基因治疗的潜在靶点。 Objective To study the differential expression of long noncoding RNA(lncRNA)in the tissues of patients with radiation-inducedrectal fibrosis(RIRF).Methods The expression of lncRNAs in rectal tissues after long term radiotherapy was detected by high throughputgene chip,and the relationship between the differential expression of lncRNAs and RIRF was analyzed.Results Hierarchical clusteringshowed that there were1622lncRNAs which was identified with differential expression more than twice fold in patients after long term radiotherapycompared with normal group,among which,1038lncRNAs were up-regulated and584lncRNAs were down-regulated(P<0.05).Intotal,76lncRNAs(54lncRNAs up-regulated and22lncRNAs down-regulated)expression were greater than tenfold difference between twogroup samples(P<0.05).Bioinformatic techniques suggested that cytokines were regulated by multiple differentially expressed lncRNAs whichare related to fibrosis signaling pathway,such as FGF2,TGF-beta1and so on.Conclusion LncRNAs may be involved in the pathogenesis ofRIRF by regulating cytokines in the fibrotic signaling pathway and it is expected to be a potential target for RIRF gene therapy.
