肺癌中热休克转录因子2促进热休克蛋白的表达 被引量:2

摘要 目的探讨HSF2通过促进热休克蛋白(heat shock protein,HSP)的表达对肺癌发生以及肿瘤细胞增殖的影响.方法选取肺癌旁组织,构建过表达HSF2真核载体(p IRES2-EGFP-HSF2),分别转染BEAS-2B、A549,检测其对细胞增殖及HSP表达的影响;转染p IRES2-EGFP-HSF2质粒的同时,共转染HSP27、HSP90的si RNA,检测其对细胞增殖的影响.结果过表达HSF2可增加BEAS-2B和A549细胞增殖速度(P<0.01),促进HSP27和HSP90的表达;转染HSP27、HSP90的si RNA均可减弱HSF2诱导的BEAS-2B和A549细胞增殖(P<0.01).结论 HSF2促进肺癌细胞增殖的作用可能主要是通过促进热休克蛋白的表达来实现. Objective To explore the influence of HSF2on occurrence of lung cancer and tumor cell proliferation by promoting HSP's expression.Methods We selected the para-carcinoma tissue from patients with lung cancer,then constructed the eukaryoticvector of HSF2overexpression,then respectively transfected into BEAS-2B and A549,in order to exam its influence on cell proliferation and HSP expression,and we co-transfected the siRNA of HSP27and HSP90while the transfection of pIRES2-EGFP-HSF2,in order to exam the influence on cell proliferation.Results The overexpression of HSF2may increase BEAS-2B,improve the A549proliferation speed(P<0.01)and the expressions of HSP27,HSP90.The co-transfected siRNA of HSP27and HSP90may equally recede the cell proliferations of BEAS-2B and A549induced by HSF2(P<0.01).Conclusion The cell proliferation of lung cancer promoted by HSF2may possibly be achieved through promoting the expression of HSP.
