葡萄糖调节蛋白94在宫颈病变中的表达

Expression of Glucose-regulated Protein 94 in Cervical Lesion

目的研究葡萄糖调节蛋白94(glucose-regulated protein94,GRP94)在宫颈病变及HPV感染中的表达及意义.方法选取2014年10月至2016年6月在昆明医科大学第二附属医院妇科门诊就诊或住院部行宫颈癌筛查(HPV、TCT),并因任一结果异常行阴道镜下活检术的患者,留取宫颈组织备用,最终根据病检结果选取正常宫颈组织40例;CIN I30例;CINII^III 30例;年龄范围为25~55岁.已行宫颈病变治疗及妊娠患者不纳入研究范围.用Western Blot法检测正常宫颈组织中以及CIN I、CINⅡ/Ⅲ组织中GRP94蛋白表达.结果正常宫颈组织中GRP94阴性、阳性表达分别占65.0%、35.0%;CINI组织中阴性、阳性分别为40.0%、60.0%;CINⅡ/Ⅲ组织中阴性、阳性分别占26.6%、73.3%.3个组阳性率总体上差异有统计学意义(P<0.05),两两比较发现:CINⅡ/Ⅲ的阳性率高于正常组,差异有统计学意义(P<0.05);CIN I的阳性率与正常组、CINⅡ/Ⅲ的阳性率相比,差异无统计学意义(P>0.05).结论 GRP94在宫颈上皮内瘤变中呈现高表达,而在正常宫颈组织中呈现低表达,GRP 94与宫颈病变进展的发生发展密切相关.

Objective To explore the expression and significance of glucose-regulated protein94in cervical lesions and HPV infection.Methods Patients from the department of gynecology of the second affiliated hospital of Kunming Medical University formOctober2014to June2016were chosen to accept cervical cancer screening(HPV and TCT),and if any result was abnormal,biopsy under colposcope was performed and the cervical tissue was collected.Finally40cases with normal cervical tissues,30cases with CINI tissues and30cases with CINII-III tissues were selected according to pathological findings,with the age range from25to55years old.The patients who had ever done cervical lesions treatment or was pregnant were not included in the study.Western Blot method was applied to detect the expression of GRP94protein in normal cervical tissues,CINI and CINII-III.Results The negative and positive expression of GRP94accounted for65.0%and35.0%respectively in normal cervical tissues,40.0%and60.0%respectively in CINI tissues,and26.6%and73.3%respectively in CINII-III tissues.There was significant difference among the overall positive rates of the three groups.Pairwise comparison showed positive rate of CINII-III group was significantly higher than that of the normal group and no significant difference was found between the positive rate of the CINI group and that of the normal group and that of CINII-III group.Conclusion GRP94shows high expression in cervical intraepithelial neoplasias,on the contrary shows lower expression in normal cervical tissue.GRP94is closely related to the occurrence of cervical lesion.
