不同抗褐变剂组合对海南龙血树组织培养及抗褐变效果的影响 被引量:1

摘要 【目的】探讨不同抗褐变剂及其组合对海南龙血树组织培养及抗褐变效果的影响,为其种苗的快速繁殖提供参考依据。【方法】采用3种抗褐变剂处理海南龙血树外植体嫩茎,开展愈伤组织诱导与抗褐变试验;在基本培养基MS中添加不同浓度的6-BA和NAA组合,检测其对海南龙血树愈伤组织分化不定芽和丛生芽形成的影响。【结果】海南龙血树嫩茎在0.1 g/L半胱氨酸(Cys)溶液中浸泡10 min后接种于含0.1 g/L维生素C(Vc)的培养基中,褐变率比对照(CK)低,仅20.5%,愈伤组织诱导率最高,为75.5%;适宜龙血树嫩茎愈伤组织诱导的植物生长调节剂组合为6.0 mg/L 6-BA+0.5 mg/L NAA,诱导率为75.5%;适宜龙血树芽增殖的植物生长调节剂组合为4.0 mg/L 6-BA+0.2 mg/L NAA,增殖倍数为3.3。【结论】采用Cys和Vc两种抗褐变剂组合处理可适当减轻龙血树组培过程中嫩茎的褐变程度,促进愈伤组织诱导;培养基中适当添加6-BA和NAA可促进愈伤组织的不定芽分化和丛生芽形成。 【Objective】Effects of different anti-browning agents and agent combinations on tissue culture and anti-browning of Dracaena cambodiana Pierre ex Gagnep.were studied to provide reference basis for rapid propagation of seedlings.【Method】Young stems of D.cambodiana explants were treated by three kinds of anti-browning agents to test the callus induction and antibrowning.Different combinations of plant growth regulator6-BA and NAA at various concentrations were added into basic medium MS to detect their effects on adventitious bud differentiation from callus and cluster bud formation of D.cambodiana.【Result】The result showed that the anti-browning rate(20.5%)was lower than CK,and callus induction rate reached the maximum(75.5%)under the treatment of soaking the young stems in0.1g/L cysteine(Cys)solution for10min and then inoculating it on medium with0.1g/L of vitamin C(Vc).Plant growth regulator combination6.0mg/L6-BA+0.5mg/L NAA was suitable for callus induction of D.cambodiana young stems,and the induction rate was75.5%.4.0mg/L6-BA+0.2mg/L NAA combination was optimal for bud proliferation,and the proliferation multiple was3.3.【Conclusion】Using anti-browning agent Cys and Vc can reduce the browning level of young stems in D.cambodiana tissue culture and improve callus induction rate.Adding appropriate amount of6-BA and NAA combinations into culture medium can promote adventitious bud differentiation from callus and cluster bud formation.
