摘要
目的探讨旋覆花素对Aβ诱导的阿尔兹海默病(AD)模型大鼠的保护作用及其作用机制。方法脑部注射Aβ25-35建立AD大鼠模型,分为模型对照组和旋覆花素给药组,另外设立空白健康对照组,每组12只大鼠。每天灌胃1次,持续给药21d。Westernblot和免疫组织化学法检测各组大鼠海马组织COX-2和iNOS的表达情况。采用Morris水迷宫法测试大鼠空间学习记忆能力。结果AD模型组大鼠海马组织中COX-2和iNOS的表达明显,较对照组大鼠增多(P<0.05)。但是给予旋覆花素治疗后,AD大鼠海马组织中COX-2和iNOS的表达受到抑制,与对照组大鼠比较差异无统计学意义;但是低于模型组大鼠(P<0.05)。旋覆花素还以改善AD模型大鼠海马组织异常形态学变化。另外,旋覆花素可以改善受试组大鼠的学习记忆能力,平均潜伏期缩短,最后一次跨平台次数增加,与对照组大鼠比较差异无统计学意义;但均优于模型组大鼠(P<0.05)。结论旋覆花素可以抑制AD大鼠海马组织COX-2和iNOS的表达,抑制海马炎症反应。
Objective To discuss the effects and mechanisms of inulicin on Alzheimer's disease(AD)model rats.Methods The rat models with AD,which were built by injecting Aβ25-35,were divided into the model control group and inulicin-treated group.Besides,there were12healthy rats in the control group.All rats were received inulicin or saline once1day for21days.The expressions of COX-2and iNOS in hippocampus tissue were measured by western blot and immunohistochemistry staining.Besides,the effects of inulicin on rats'learning and memory were evaluated by Morris water maze method.Results After treatment,the expressions of COX-2and iNOS in hippocampus tissue of AD model rats were higher than the control rats(P<0.05).But the the expressions of COX-2and iNOS in hippocampus tissue of AD model rats with inulicin were inhibited,which were lower than the model rats(P<0.05).The abnormal histomorphology of AD rats'hippocampus was improved by inulicin.Besides,inulicin improved the learning and memory capacity in the inulicin treated rats,which were with less incubation and more times of acrossing the target area than the AD model rats,(P<0.05).Conclusion There are significant evidences that inulicin could effectively inhibit the expressions of COX-2and iNOS of AD rats'hippocampus,and decrease the inflammation,which would have a good prospect of clinical application for the treatment of Alzheimer's disease.
