注射用心肌肽对幼鼠离体心脏缺血再灌注能量代谢的影响

摘要 目的分析注射用心肌肽对幼鼠离体心脏缺血再灌注能量代谢的影响。方法选取选取60只SD幼鼠,随机分为5组,每组各为12只。正常对照组中幼鼠进行心脏灌流20min后,持续灌注150min。另一组在正常对照组基础上注射50mg/L心肌肽。模型对照组幼鼠进行心脏灌流20min后,停博液缺血90min,恢复灌注1h,建立心肌缺血再灌注损伤模型,应用K-H缓冲液进行灌流。CMP1(注射用心肌肽1组)幼鼠在停搏液加入100mg/L注射心肌肽。CMP2(注射用心肌肽2组)幼鼠在停搏液加入100mg/L注射心肌肽,灌流液加入注射用50mg/L心肌肽。比较分析5组幼鼠相关临床指标对比。结果正常对照组幼鼠离体心脏缺血再灌注后,心功能并无显著变化,应用注射用心肌肽后,心脏功能并无显著影响。CMP2幼鼠再灌注1h后,心率为(188.20±15.44)次/min显著多于CMP1组(164.30±15.66)次/min(P<0.05)。CMP2幼鼠再灌注1h后,冠状动脉流量显著优于CMP1组(P<0.05)。用药组心肌结构相对完整度较高,SOD活性显著高于模型对照组(P<0.05)。结论注射用心肌肽可显著改善幼鼠离体心脏缺血再灌注能量代谢,保护心肌功能和再灌注损伤,大大提高冠状动脉流量。 Objective To analyze cardiomyopeptide for injection rat isolated heart ischemia effect of reperfusion on energy metabolism.Methods60SD rats were selected and randomly divided into5groups,12rats in each group.In the normal control group,the rats were perfused for20minutes and perfused for150minutes.The other group was injected with50mg/L myocardial peptide on the basis of normal control group.In the model control group,the rats were perfused with K-H for20minutes,then stopped the fluid for90minutes,then resumed the perfusion of1hour,then established the model of myocardial ischemia reperfusion injury,and then carried out the perfusion with the buffer.CMP1(cardiomyopeptide for injection1group)rats were injected with100mg/L myocardial peptide in the cardioplegic solution.CMP2(cardiomyopeptide for injection2group)were injected with100mg/L myocardial peptide in the cardioplegic solution,and the injected50mg/L myocardial peptide was injected into the perfusate.Comparison and analysis of relative clinical indexes of several groups of young rats.Results The rats in the normal control group isolated heart ischemia reperfusion injury,heart function no significant changes,the application of cardiomyopeptide for injection,no significant effect on cardiac function.After reperfusion with1hour in CMP2rats,the heart rate was(188.20±15.44)times per min significantly more than(164.30±15.66)times per min in the CMP1group(P<0.05).After reperfusion with1hour in CMP2rats,the coronary flow rate were significantly better than those in group CMP1(P<0.05).The myocardial structural integrity of the treatment group was relatively high,and the activity of SOD was significantly higher than that of the model control group(P<0.05).Conclusion Cardiomyopeptide for injection can significantly improve the myocardial ischemia reperfusion energy metabolism in neonatal rats,protect myocardial function and reperfusion injury,and greatly increase the coronary flow.
