摘要
目的探讨人参皂苷rh2在SD大鼠口腔白斑癌变过程中的抑制作用以及对血管内皮生长因子(VEGF)表达的影响。方法通过7,12-二甲基苯并[a]蒽(DMBA)诱导建立SD大鼠颊部白斑动态模型30例,随机分成2组:A组(生理盐水组)15例、B组(人参皂苷rh2组)15例,于4、7、10周处死后取双侧颊粘膜组织进行病理学检测和免疫组织化学染色,并进行卡方检验和秩和检验统计分析。结果两组癌变过程中不同组织学分布有显著差异(P<0.05),B组病变程度明显轻于A组。两组中VEGF表达均随着病变的严重程度而加强。A、B两组相同病损进行VEGF阳性率比较结果,单纯增生组、轻度增生组、中度增生组均无统计学意义(P>0.05)。结论人参皂苷rh2对大鼠口腔白斑癌变有抑制作用,但是其对颊粘膜上皮内VEGF的表达影响不大。
Objective To explore the inhibitory effect of Ginsenoside Rh2on the carcinogenesis of oral leukoplakia in SD rats and the effect of vascular endothelial growth factor(VEGF)expression.Methods30dynamic models of oral leukoplakia in SD rats were induced by7,12-dimethyl benzene and[a]anthracene(DMBA)and randomly divided into two groups.A group was treated with normal saline,B group was treated with Ginsenoside Rh2.Bilateral buccal mucosal tissue was used for pathological examination and immunohistochemical staining after the execution on4th,7th and10th week and then the data was analysed with chi-square test and rank sum test.Results The occurrence of two groups histology difference was signifi cant in the development of oral cancer(P<0.05),group B was signifi cantly less pathological than group A.the expression of VEGF in both groups gradually strengthened with the severity of the lesion.The VEGF positive rate was compared between the same lesion in two groups:there was no significant difference on the simple hyperplasia,the mild dysplasia and the moderate dysplasia(P>0.05).Conclusion There was a inhibition of Ginsenoside Rh2on the carcinogenesis of oral leukoplakia in rats,but it had little effect on the expression of VEGF in the buccal mucosal epithelium.
