黄芪多糖对乳腺癌细胞抑制作用实验研究 被引量:11

摘要 目的:探讨黄芪多糖对乳腺癌细胞生长的抑制作用。方法:应用MTT法和克隆形成法检测黄芪多糖对人乳腺癌MCF-7细胞的生长抑制作用,流式细胞术检测黄芪多糖对人乳腺癌MCF-7细胞的细胞周期的影响。结果:不同药物浓度的黄芪多糖作用于MCF-7后的细胞克隆数组间差异有统计学意义(P<0.05);随黄芪多糖浓度的增大MCF-7细胞的存活率出现明显降低,3 mg/mL的黄芪多糖对MCF-7细胞的抑制作用随时间延长而增强。黄芪多糖产生作用后,S期细胞比例减少,而G0/G1期细胞比例增加。结论:黄芪多糖对人乳腺癌细胞有显著的生长抑制作用,这种作用随药物浓度和时间的增加而加强,诱导细胞周期阻滞于G0/G1期可能是其机制之一。 Objective:To investigate the inhibitory effect of astragalus polysaccharides on the growth of breast cancer cells.Methods:MTT method and clone formation method were used to detect the inhibitory effect of astragalus polysaccharides on human breast cancer MCF-7cells,and the effect of astragalus polysaccharides on the cell cycle of human breast cancer MCF-7cells was detected by flow cytometry.Results:There were significant differences in MCF-7cell clone array after the interaction between the different concentration of astragalus polysaccharide(P<0.05).With the increase of the concentration of astragalus polysaccharide on MCF-7cell survival rate decreased significantly,3mg/mL inhibitory effect of astragalus polysaccharides on MCF-7cells increased with time.After the action of astragalus polysaccharide,the percentage of S phase cells decreased,while the percentage of G0/G1phase cells increased.Conclusion:Astragalus polysaccharide has significant inhibitory effect on human breast cancer cells,which is enhanced with the increase of drug concentration and time,and the induction of cell cycle arrest in G0/G1phase may be one of the mechanisms.
