调经汤对卵巢早衰模型大鼠卵巢功能影响的实验研究 被引量:10

摘要 目的观察调经汤对卵巢早衰模型大鼠卵巢功能的影响。方法将60只雌性成熟未交配Wistar大鼠随机分为空白对照组、模型组、阳性药物对照组、调经汤低、中、高剂量组,每组10只。空白组大鼠按0.4mL/100g体质量腹腔注射无菌注射用水,其余各组大鼠按0.4mL/100g体质量腹腔注射浓度为0.5mg/mL的顺铂溶液,连续7天。观察大鼠行为变化、动情周期,采用放射免疫法检测大鼠血清促卵泡生成素(FSH)、雌激素(E2)和促黄体生成素(LH)水平,光镜观察卵巢组织形态改变。结果模型组和阳性药物对照组大鼠无稳定的动情周期,调经汤中、高剂量组逐渐恢复规律的动情周期。给药后第7、14天,与空白对照组比较,模型组大鼠血清FSH、LH升高,E2降低,差异有统计学意义[第7天:(7.38±0.99)IU/L比(6.18±0.24)IU/L,(3.35±0.27)ng/L比(2.79±0.24)ng/L,(94.04±6.22)ng/L比(109.10±5.69)ng/L,P均<0.05;第14天:(8.01±0.13)IU/L比(6.97±0.88)IU/L,(4.22±0.82)ng/L比(3.20±0.17)ng/L,(131.74±23.65)ng/L比(157.82±5.68)ng/L,P均<0.05];给药后第7天,与模型组比较,调经汤低、中、高剂量组大鼠血清FSH降低,E2升高,差异有统计学意义[(5.72±0.51)IU/L、(6.20±1.05)IU/L、(6.08±0.26)IU/L比(7.38±0.99)IU/L;(104.46±10.25)ng/L,(104.48±12.85)ng/L,(106.64±8.67)ng/L比(94.04±6.22)ng/L,P均<0.05];血清LH含量水平虽降低,但差异无统计学意义[(2.96±0.38)ng/L、(3.00±0.19)ng/L、(2.96±0.23)ng/L比(3.35±0.27)ng/L,P>0.05]。给药后第14天,与模型组比较,调经汤中剂量组大鼠血清FSH、LH降低,E2升高,差异有统计学意义[(7.02±1.04)IU/L比(8.01±0.13)IU/L,(3.37±0.53)ng/L比(4.22±0.82)ng/L,(153.67±20.55)ng/L比(131.74±23.65)ng/L,P均<0.05]。模型组大鼠卵巢萎缩,卵泡生长少,闭锁卵泡增加,部分被纤维组织代替,黄体数量少,子宫内膜明显变薄。调经汤低、中、高剂量组大鼠卵泡数量和黄体数目不同程度增多,子宫内膜有所增厚。结论调经汤能有效改善顺铂诱导的卵巢早衰模型大鼠的卵巢功能。 Objective To investigate the effect of Tiaojing decoction on treatment of premature ovarian failure in rats.Methods Sixty female adult rats were randomly divided into blank group,model group,positive drug control group,and low-,medium-,and high-dose of Tiaojing decoction groups,with10rats in each group.Premature ovarian failure rat model was established through7-consecutive-day intraperitoneal injection of cisplatin solution in all groups except for blank group.The oestrous cycle was observed,the variation in content of FSH,E2,LH was detected with radioimmunoassay and ovarian pathological changes were observed under light microscope in all group.Results The model group and positive drug control had no stable estrous cycle while high-dose and medium dose Tiaojing decoction group gradually restored the estrous cycle.On day7and14,the serum FSH(Day7:7.38±0.99IU/L,Day14:8.01±0.13IU/L)and LH(Day7:3.35±0.27)ng/L,Day14:4.22±0.82ng/L)increased and E2(Day7:94.04±6.22ng/L,Day14:131.74±23.65ng/L)decreased compared with those in blank group(FSH:Day7:6.18±0.24IU/L,Day14:6.97±0.88IU/L;LH:Day7:2.79±0.24ng/L,Day14:3.20±0.17ng/L;E2:Day7:109.10依5.69ng/L,Day14:157.82±5.68ng/L),all with P<0.05;on day7,compared with those in the model group,the serum FSH content decreased and E2content increased in the low-,middle-and high-dose Tiaojing decoction groups(FSH:5.72±0.51IU/L,6.20±1.05IU/L,6.08±0.26IU/L;E2:104.46±10.25ng/L,104.48±12.85ng/L,106.64±8.67ng/L,all P<0.05),but the serum LH level decreased without significant difference(2.96±0.38ng/L,3.00±0.19ng/L,2.96±0.23ng/L,P>0.05);on day14,the FSH(7.02±1.04IU/L)and LH(3.37±0.53ng/L)in the middledose of Tiaojing decoction group were lower and E2(153.67±20.55ng/L)was higher than those in the model group(P<0.05).Ovarian tissue pathological changes such as ovarian volume contraction,less follicular growth,increase in atresia follicles,partly replaced by fibrous tissue,less corpus luteum in number,and thinner endometrium were observed in the model group.These pathological changes were improved in Tiaojing decoction groups,such as more follicular growth,increase in corpus luteum number,and thicker endometrium.Conclusion Tiaojing decoction can improve the ovarian function of rats with premature ovarian failure induced by cisplatin.
