ClassⅠ/Ⅱ新城疫弱毒株诱导SPF鸡免疫损伤及黏膜免疫应答的比较分析 被引量:2

摘要 为了比较Class Ⅰ新城疫弱毒JS10株和Class Ⅱ新城疫弱毒La Sota株免疫雏鸡后的免疫损伤和黏膜应答情况。本研究将60只SPF白来航鸡,随机分成A、B、C 3组。7日龄时,A组和B组分别用JS10株和La Sota株进行滴鼻、点眼免疫,C组以同样方式接种PBS,并于免疫后的d1、4、7、14,每组随机抽取5只扑杀,分别收集唾液、气管、肺和肠的冲洗液,使用双夹心ELISA试剂盒测定SIgA、IgM和IgY的抗体水平。结果显示,2株病毒均未产生免疫损伤;JS10株产生的黏膜抗体水平均高于La Sota株,并且免疫后的d7、14的唾液、气管和肠冲洗液中的SIgA的抗体水平以及肺中IgY抗体水平均显著高于La Sota(p<0.05)。研究表明,2株新城疫弱毒滴鼻、点眼后都能诱导消化道和呼吸道产生强烈的黏膜免疫反应,并且与免疫后的日龄存在明显相关性。由于JS10株可诱导更好的黏膜免疫应答,提示其可作为新型新城疫弱毒疫苗的候选株。 The objective of the present study was to investigate the immunological injury and mucosal response induced by attenuated Newcastle disease virus strains JS10(Class I)and La Sota(Class II).In this experiment,60SPF chicks were randomly divided into group A,B and C.At the age of7days,chicks in group A and B were respectively immunized with JS10strain and La Sota strain and chicks in group C received PBS as controls.5chicks in each group were randomly terminated at d1,4,7or14post immunization.Saliva,trachea,lung and intestinal samples were collected for detection of mucosal antibodies SIg A,Ig M and Ig Y using double sandwich ELISA kits.The results showed that these two NDV strains did not cause any immune injury.The JS10strain induced higher levels of mucosal antibodies as compared with La Sota strain and significant differences(p<0.05)were revealed at d7and d14post immunization.
