摘要
目的建立视神经脊髓炎转基因(2D2)小鼠的繁育方法,并对其子代鼠进行基因鉴定。方法将雌雄2D2转基因小鼠各1只与C57BL/6野生型小鼠(1∶1)进行交配,由鼠尾血提取基因组DNA,应用PCR技术扩增,电泳后观察2D2转基因传代小鼠的基因表型。结果共繁育产生56只子代小鼠,其中36只子代小鼠基因组DNA扩增出657bp的2D2条带,阳性率为64.3%。结论成功利用杂交方式有效繁殖2D2转基因鼠和应用PCR技术进行基因鉴定,为后续视神经脊髓炎的实验研究奠定了基础。
