低分子肝素对人肺腺癌细胞株A549增殖及VEGF、TGF-β1表达的影响

摘要 目的研究低分子肝素(LMWH)干预体外培养的人肺腺癌细胞株A549后对其增殖抑制及血管内皮生长因子(VEGF)、转化生长因子β1(TGF-β1)表达的影响。方法采用四甲基偶氮唑盐(MTT)法分别检测对照组(未加入LMWH)和实验组(LMWH干预的质量浓度分别为0.5、1.0、2.0、5.0、10.0μg/mL)干预24、48、72 h后的吸光度值,并计算增殖抑制率;采用酶联免疫吸附试验(ELISA)检测LMWH干预A549细胞72 h后上清液中VEGF及TGF-β1表达水平,并对VEGF和TGF-β1进行相关性分析。结果 LMWH干预24 h时,对照组与实验组积分光密度(IOD)值比较,差异无统计学意义(P>0.05);LMWH干预48、72 h时,实验组IOD值均较对照组低,差异均有统计学意义(P<0.05);LMWH干预72 h后,实验组VEGF、TGF-β1表达水平均下降,差异均有统计学意义(P<0.05),且二者呈显著正相关(r=0.934,P<0.05)。结论LMWH可以抑制体外培养的人肺腺癌A549细胞的增殖,且该抑制作用呈质量浓度和时间依赖性。其机制可能与下调肺癌A549细胞VEGF和TGF-β1的表达有关。 Objective〓To study the effects of low molecular weight heparin(LMWH)intervention on the proliferation of in vivo cultured human lung adenocarcinoma A549cell line and expression of VEGF and TGF-β1.Methods〓The MTT method was adopted to detect the IDO value in the control group(without adding LMWH)and experimental groups(LMWH intervention mass concentrations0.5,1.0,2.0,5.0,10.0μg/mL)after24,48,72h intervention.The proliferation inhibition rate was calculated.The levels of supernatant VEGF and TGF?鄄β1after LMWH intervention for72h on A549cells were detected by using the ELISA method.Then the correlation analysis on VEGF and TGF?鄄β1was conducted.Results〓After24h intervention of LMWH,the IOD value had no statistical difference between the control group and experimental groups,the difference was not statistically significant(P>0.05);at48,72h intervention of LMWH,the IOD value in the experimental groups were lower than that in the control group and the difference has statistical significance(P<0.05);after72h intervention of LMWH,the expression levels of VEGF and TGF?鄄β1in the experimental groups were declined,the difference was statistically significant(P<0.05),moreover both showed significantly positive correlation(r=0.934,P<0.05).Conclusion〓LMWH could inhibit the proliferation of in vitro cultured human lung adenocarcinoma A549cells,moreover the inhibiting effect shows the concentration and time dependence,its mechanism may be related with down-regulating the expression of VEGF and TGF?鄄β1in lung adenocarcinoma A549cells.
