摘要
目的探究麦冬皂苷B(ophiopogonin-B,OP-B)对非小细胞肺癌细胞株H460有丝分裂及细胞周期调控的相关分子机制。方法 TUNEL免疫组化法观察细胞核物质的变化;DAPI染色法观察细胞核形态的改变及有丝分裂的状态;Western blot法检测细胞周期调控相关的蛋白信号通路。结果形态学结果表明,10μmol·L^(-1)浓度的OP-B能明显抑制H460细胞的有丝分裂,蛋白水平的检测结果表明,OP-B能抑制cyclin D1、cyclin B1的表达,促进Myt的表达及Cdc2的磷酸化,抑制histone H3(Ser10)的磷酸化,同时能上调周期抑制性蛋白p21的表达。结论 OP-B通过Myt/Cdc2信号通路导致细胞周期停滞在G0/G_1期,从而抑制细胞的有丝分裂。
AimTo explore the effects of ophiopogonin B(OP B)on cell cycle and mitosis in non small cell lung cancer(NSCLC)H460cells in vitro and the underlying mechanisms.MethodsTUNEL immunohistochemical assay was used to detect the change of the nuclear matter.DAPI staining was used to detect the change of the nuclear morphology and the mitosis status.Meanwhile,Western blot was performed to determine the protein level of the proteins regulating cell cycle and mitosis.ResultsOP B significantly arrested cell cycle in G0/G1phase and inhibited the mitosis in H460cells at the concentration of10μmol·L-1.Meanwhile,it inhibited the protein level of cyclinD1,cyclinB1,and up regulated the expression of Myt and the phosphorylation level of Cdc2.ConclusionOP B inhibits cell mitosis in A549cells through Myt/Cdc2signaling pathway.
