白藜芦醇对过氧化氢诱导巨噬细胞炎性衰老的抑制作用及机制 被引量:1

摘要 目的观察白藜芦醇(Res)对过氧化氢(H_2O_2)诱导的THP-1巨噬细胞炎性衰老的抑制作用,并探讨其作用机制。方法用160 nmol/L佛波酯诱导人单核细胞THP-1巨噬细胞,将其随机分为四组,空白对照组给予培养基培养;H_2O_2组给予培养基培养1 h后再加300μmol/L H_2O_2培养24 h;Res+H_2O_2组给予40μmol/L Res培养1 h,再加300μmol/L H_2O_2培养24 h;Res组给予40μmol/L Res培养24 h。用衰老相关β-半乳糖苷酶染色法检测衰老THP-1巨噬细胞比例,用Western blotting法检测THP-1巨噬细胞内视网膜细胞瘤蛋白(Rb)、磷酸化视网膜细胞瘤蛋白(pRb)、自噬微管相关蛋白1清链3(LC3)、Bcl-2同源结构域蛋白(Beclin1)、泛素结合蛋白(p62)表达;ELISA法检测THP-1巨噬细胞培养液中TNF-α、IL-1β、IL-6水平。结果与空白对照组比较,H_2O_2组衰老细胞比例高(P<0.05);与H_2O_2组比较,Res+H_2O_2组衰老细胞比例低(P<0.05)。与空白对照组比较,H_2O_2组Rb蛋白表达高、p-Rb蛋白表达低(P均<0.05);与H_2O_2组比较,H_2O_2+Res组Rb蛋白表达低、p-Rb蛋白表达高(P均<0.05)。与空白对照组比较,H_2O_2组,LC3-Ⅱ、Beclin-1蛋白表达及LC3-Ⅱ/LC3-Ⅰ低,p62蛋白表达高(P均<0.05);与H_2O_2组比较,H_2O_2+Res组LC3-Ⅱ、Beclin-1蛋白表达及LC3-Ⅱ/LC3-Ⅰ高,p62蛋白表达低(P均<0.05)。与空白对照组比较,H_2O_2组TNF-α、IL-1β、IL-6水平高(P均<0.05);与H_2O_2组比较,H_2O_2+Res组TNF-α、IL-1β、IL-6水平低(P均<0.05)。空白对照组与Res组以上指标比较,P均>0.05。结论白藜芦醇可能通过促进细胞自噬来抑制H_2O_2诱导的THP-1巨噬细胞炎性衰老。
