多叶勾儿茶主要成分提取及抗氧化活性初探 被引量:5

Extraction and Antioxidant Activity of Major Components from Berchemia polyphylla

摘要 为探究多叶勾儿茶提取芦丁和槲皮素的最佳工艺及其体外抗氧化活性,为该中药材的开发和利用提供理论依据,采用回流提取法和响应面法,考察乙醇体积分数、温度、料液比3个因素及其交互作用和时间对提取工艺的影响,通过粗提物还原力和对·OH的清除能力考察其体外抗氧化活性。结果表明:多叶勾儿茶芦丁和槲皮素的最佳提取条件为乙醇体积分数73.86%,温度61.96℃,液料比36.28mL/g,提取时间为3h,此条件下芦丁、槲皮素含量分别为15.45 mg/g和0.15 mg/g,与实际值误差小,粗提物浓度为0.08~0.1mg/mL时还原力强,0.6~1.0mg/mL时对·OH清除率较高。该提取工艺稳定合理,多叶勾儿茶粗提物具有较好的抗氧化能力。 The effect of three factors(ethanol concentration,liquid/solid ratio and temperature),different interactions(liquid/solid ratio×temperature,ethanol concentration×temperature and liquid/solid ratio × ethanol concentration)and extraction time on rutin and quercetin extraction from B.polyphyllais analyzed by the reflux extraction and response surface method and the in vitro antioxidant activity of the crude extract is determined by analyzing reducing activity and·OH scavenging rate of the crude extract to discuss the optimum extraction technology of rutin and quercetin extraction from Berchemia polyphylla and the in vitro antioxidant activity.Results:The optimum extraction conditions for rutin and quercetin extraction from B.polyphyllainclude 73.86% ethanol concentration,36.28 mL/g liquid/solid ratio,3 hextraction time and 61.96℃.The extraction amount of rutin and quercetin form B.polyphylla under the optimum extraction conditions is 15.45 mg/g and 0.15 mg/g respectively,which is no obvious difference with the actual value.The reducing activity of the crude extract with 0.08~0.1 mg/mL is high and the·OH scavenging rate of the crude extract with 0.6~1.0 mg/mL is relatively high.The extraction technology with advantages of good stability and rationality and better antioxidant capacity of the crude extract from Berchemia polyphylla provide the theoretical basis for development and utilization of Chinese herbal medicines.
