摘要
为解决鸡枞菌分离较难的问题,对采自秦岭地区的一株野生鸡枞菌,采用组织分离法对标本菌株进行分离纯化,通过平皿观察法和ITS序列分析确定其菌属,以菌丝生长速率为指标、探索其最优生长碳源、氮源、KH_2PO_4浓度、MgSO_4浓度、温度及酸碱度等培养条件。结果表明:经形态特征及ITS序列分析确定此菌株为鸡枞菌,该菌营养菌丝生长最适碳源是玉米粉;最适氮源是硝酸铵;鸡枞菌生长较快的KH_2PO_4浓度在0.10%~0.20%时,最适浓度是0.15%;MgSO_4浓度在0.025%~0.075%时,最适浓度是0.050%;生长温度范围在25~27℃时,最适温度是26℃;pH范围5.0~6.0,最适pH是5.0。
To solve the difficult problem of separating Termitomyces albuminosus,the isolation,identification and growth characteristics of Qinling Mountains area from a wild Termitomyces albuminosus were studied,tissue isolation method was used to isolate and purify the strains,its genus was determined by plate observation and ITS sequence analysis,the optimum growth carbon source,nitrogen source,KH2PO4concentration,MgSO4concentration,temperature and pH were investigated with the growth rate of mycelium as index.The results showed that according to the morphological characteristics and ITS sequence analysis to determine the strain of Termitomyces albuminosus,the optimum carbon source for the growth of mycelia was corn flour;the optimum nitrogen source was ammonium nitrate;the concentration of KH2PO4was0.10%~0.20%,the optimum concentration was0.15%;the concentration of MgSO4was0.025%~0.075%,the optimum concentration was0.050%;the growth temperature range was25~27℃,the optimum temperature was26℃;the growth pH range was5.0~6.0,the optimum pH was5.0.
