摘要
目的探索急性脊髓损伤后大鼠脊髓组织中c-Jun氨基末端激酶(c-Jun amino-terminal kinase,JNK)基因表达与神经功能修复状态的关联。方法取健康成年雄性大鼠50只,随机选取,假手术对照组18只,实验组32只。损伤后1、12、24、72 h行PCR检测,比较大鼠脊髓组织中JNK(JNK1、JNK2、JNK3)表达水平变化;损伤后12、72 h选取L_(4/5)节段脊髓行HE染色、,比较脊髓损伤大鼠脊髓组织细胞凋亡情况、神经功能变化状态(NSS)。结果 PCR结果显示,髓损伤大鼠脊髓组织中JNK1、JNK2、JNK3的表达,在损伤后1、12、24、72 h除JNK3在相应时点表达水平与假手术组比较水平下降外,其他两项表达与假手术对照组无明显差别。L4/5脊髓HE染色结果提示,与假手术组比较,损伤后12 h损伤的脊髓皮质周边区HE染色阳性细胞显著增多。随着时间推移,染色阳性细胞出现明显减少的趋势,损伤后12 h的染色阳性细胞明显少于72 h。损伤组NSS评分则明显更高,但脊髓损伤组损伤后不同时点比较,NSS评分渐减。结论脊髓损伤后出现JNK3表达下调的情况,可能与神经细胞凋亡以及促进损伤后神经功能的恢复有关联。
Objective To explore the association between spinal cord tissue state c-Jun N-terminal kinase(JNK)gene expression and recovery of neural function acute spinal cord injury.Methods50healthy adult male were randomly selected sham group18,a total of32experimental group.Comparing the injury spinal cord tissue JNK expression levels as well as spinal cord injury in spinal cord tissue cell apoptosis andNSS change state after injury.Results The cord injury in spinal cord tissue JNK1,JNK2,JNK3expression were examined for the comparison and found that among the groups after1,12,24,72h after the injury JNK3expression level of sham group level showed difference after injury.And in this study we selected L4/5spinal cord HE staining,the results suggested that compared with the sham group,12h after injury in cortical damage the surrounding area HE staining positive cells increased significantly.After the damage,staining positive cells was significantly reduced.Sham group of24h the NSS score showed a small increase,while in the injury group the NSS score was significantly increased;compared various time points after spinal cord injury group,the NSS scores tended to decrease.Conclusion JNK3downregulated after spinal cord injury may be associated with neuronal apoptosis,the downturn is likely to promote the recovery of nerve function after injury.
