WNT信号抑制基因SFRP5 mRNA表达与胃癌的关系

Association between mRNA expression of WNT antagonist gene SFRP5 and gastric cancer

目的为了探讨WNT信号通路抑制基因SFRP5mRNA表达水平与胃癌发生及临床病理之间的关系。方法课题组前期收集了35例胃癌及癌旁样本,通过real-time PCR(Taqman探针法)检测该基因mRNA表达水平,利用2^(-ΔCt)及2^(-ΔΔCt)公式对表达数据进行处理,然后将表达数据与临床信息进行据统计分析。结果最终研究发现:(1)SFRP5基因在胃癌组织中表达水平较癌旁组织明显降低(P=0.000 2),说明该基因的表达下调,在胃癌发生中起着重要作用;(2)表达数据与临床信息统计后发现该基因的表达水平与患者的淋巴结转移密切相关。结论 SFRP5基因的异常表达在胃癌进展中可能也起着重要作用。

Objective In the present study,we investigated the mRNA expression of WNT antagonist gene SFRP5and its regulatory role in gastric cancer and the clinicopathologic characteristics.Methods The quantitative mRNA expression level was determined by real-time PCR in35frozen samples of gastric cancer and paired adjacent non-cancerous tissue.The relative amount of target RNA was calculated using the equation2Ct and2Ct.Results Statistical analysis showed that the mRNA expression of SFRP5was significantly lower in gastric cancer than in non-cancerous tissues,which suggested this gene plays an important role in gastric cancer.Conclusions The level of SFRP5expression was significantly associated with Lymph node metastasis,which indicated that the expression of SFRP5may also play a key role in gastric cancer progression.
