NSCLC组织中FOXP1和FOXQ1的表达及相关性研究 被引量:4

摘要 目的:探讨叉头框蛋白P1(forkhead box P1,FOXP1)和叉头框蛋白Q1(forkhead box Q1,FOXQ1)在非小细胞肺癌(non-small cell lung cancer,NSCLC)组织中的表达与临床病理特点、预后的相关性,以及二者是否存在相互关系、二者联合检测在NSCLC中的意义。方法:采用免疫组化En Vision两步法检测207例NSCLC组织及相对应的癌周正常肺组织中FOXP1和FOXQ1蛋白的表达。使用卡方检验和Spearman's rank检验分析二者在NSCLC中表达的相关性。为了评估联合检测FOXP1和FOXQ1对预后的意义,将207例NSCLC组织再分为A组(FOXP1低表达/FOXQ1高表达)(100例)和B组(非FOXP1低表达/FOXQ1高表达)(107例)。结果:FOXP1表达率与性别、淋巴结转移、临床分期、病理类型有关(P<0.05)。FOXP1低表达组的患者在NSCLC中生存时间较短,即预后差。FOXQ1表达率与淋巴结转移、临床分期有关(P<0.05)。FOXQ1高表达组的患者在NSCLC中生存时间较短,即预后差。生存曲线提示A组总体生存时间明显比B组生存时间要短。卡方检验揭示了二者之间存在着负相关性(P<0.001)。多因素分析结果显示低表达的FOXP1、高表达的FOXQ1、FOXP1低表达/FOXQ1高表达、淋巴结转移分期、临床分期是NSCLC的独立预后因素。结论:FOXP1和FOXQ1参与了NSCLC的发生发展及转移,且与预后有关。二者具有负相关性,联合检测FOXP1和FOXQ1能更好地判断NSCLC患者的预后,可作为NSCLC诊断及判断预后的联合指标之一。 Objective:Forkhead box P1(FOXP1)and forkhead box Q1(FOXQ1)expression in non-small cell lung cancer(NSCLC),together with their isolated predictive values regarding patient prognosis,have been evaluated in our previous studies.However,it remains unknown as to whether or not there is a correlation between the two markers;their combined expression in combination with clinical and pathologic variables in NSCLC,as well as the prognostic value of FOXP1combined with FOXQ1expression,have not been explored.Therefore,the aim of this study was to investigate these issues.Methods:Immunohistochemistry was performed to assess the expression of FOXP1and FOXQ1in207patients with NSCLC and in207paracancerous(paratumorous)normal lung tissues.The correlations between the immunohistochemical expression of FOXP1and FOXQ1in NSCLC were examined using either theχ2test and Spearman’s rank test.Additionally,we reclassified patients into two groups:FOXP1Low/FOXQ1High(100patients)and non-FOXP1Low/FOXQ1High(107patients).Results:The expression of FOXP1in NSCLC was related to gender,lymph node metastasis,clinical stage,pathological type(P<0.05).Patients with low FOXP1expression have a shorter survival time in NSCLC,which means poor prognosis.The expression of FOXQ1in NSCLC was related to lymph node metastasis,clinical stage(P<0.05).Patients with high FOXQ1expression have a shorter survival time in NSCLC,that is poor prognosis.Survival curves suggest that the overall survival time of group A was significantly shorter than that of group B.Chi-square test revealed a negative correlation between the two factors(P<0.001).Multivariate analysis showed that low expression of FOXP1,high expression of FOXQ1,FOXP1Low/FOXQ1High,lymph node metastasis stage,clinical stage are independent prognostic factor in NSCLC.Conclusion:FOXP1and FOXQ1are involved in the development and metastasis of NSCLC,also are related to the prognosis.A strong negative correlation between FOXP1and FOXQ1expression(P<0.001)was observed,combined detection of FOXP1and FOXQ1can better judge the prognosis of patients with NSCLC,which can be used as one of the combined indicators of NSCLC diagnosis and prognosis.
