马钱子水煎液对肺癌细胞体内外的抑制作用

The Inhibiting Effect of nux vomica decoction on lung cancer cells in vitro and in vivo

[目的]研究马钱子水煎液在体内和体外对肺癌细胞的作用。[方法]体外实验:用MTT和划痕试验检测马钱子水煎液对Lewis肺癌细胞的增殖和迁移能力的影响。体内实验:建立小鼠Lewis细胞气管滴注肺癌模型,给予马钱子水煎液0.1 g/kg进行灌胃治疗,1次/d,给药期间间隔4 d记录1次小鼠的生存状态,连续7周,处死小鼠后HE染色法进一步评价马钱子水煎液对肺部组织病理变化。[结果]体外实验(MTT实验)表明,马钱子水煎液有抗肺癌细胞增殖作用并呈剂量依赖性,划痕试验结果说明马钱子水煎液抑制其迁移的作用并呈时间依赖性;体内实验结果表明,给药组生存质量相比模型组有显著提;HE染色结果显示,给药组未见明显的细胞核增多、增大。[结论]马钱子水煎液能有效抑制体外肿瘤细胞增殖和迁移,并且具有体内抗肿瘤作用。

〔Objective〕To research the effect of nux vomica decoction on lung cancer cells in vitro and in vivo.〔Methods〕In vitro:In this study,MTT and cell scratch were used to determine the effect of nux vomica on cell proliferation and migration in mice lewis lung cancer cells.In vivo:Orthotopic lewis lung cancer model in mice was established by injecting lewis lung cancer cells into trachea.The mice received nux vomica decoction[0.1g/kg per mouse]via intragastric administration once daily until all mice were scarified at seven weeks later.The survival status of the mice was recorded once every four days.The effect of nux vomica decoction on pathological changes of pulmonary was further investigated by HE staining.〔Results〕In vitro:The MTT experiments results revealed that nux vomica decoction could be suppress proliferation in a dose dependent matter.And nux vomica decoction markedly reduced cells migration in a time dependent matter,as indicated by cell scratch.In vivo revealed that nux vomica decoction could improve life quality.The HE staining results showed that there were relatively few dense nuclei in the treated mice with nux vomica decoction.〔Conclusion〕nux vomica decoction could effectively inhibit the proliferation and migration of tumor cells in vitro and resist tumor in vivo.
