摘要 目的探讨miR-21通过Wnt信号通路影响前列腺癌的生物学行为的机制。方法应用q PCR检测前列腺癌DU145、PC-3和LNCa P细胞株中miR-21 mRNA的表达水平。通过转染miR-21-mimic模拟物和miR-21-inhibitor模拟物上调和下调DU145细胞中miR-21的表达。细胞凋亡实验检测上调miR-21表达对DU145细胞凋亡行为的影响;划痕愈合实验检测上调miR-21表达对DU145细胞迁移能力的影响;Western blotting检测上调miR-21表达与Wnt信号通路蛋白表达的关系。结果 DU145细胞株miR-21 mRNA表达水平明显高于PC-3和LNCa P细胞株(P<0.05)。上调miR-21表达可以抑制DU145细胞的凋亡,增强DU145细胞的迁移能力,还可以上调Wnt信号通路蛋白的表达水平(P<0.05)。结论 miR-21可能通过Wnt信号通路影响前列腺癌的生物学行为。 Objective To investigate affecting mechanism of miR-21on biological behavior of prostate cancer by Wnt signaling pathway.Methods The miR-21and mRNA expression levels in prostate cancer DU145,PC-3and LNCaP cell lines were detected by quantitative polumerase chain reaction(qPCR).The miR-21expression in DU145cells was up and down regulated by transfecting miR-21mimic mimics and miR-21inhibitor mimics.Effect of up regulated miR-21expression on apoptosis behavior of DU145apoptosis was detected by apoptosis experiment;effect of up regulated miR-21expression on ability of DU145cell migration by scarification healing experiment;relationship between up regulated miR-21expression and protein expression of Wnt signaling pathway was detected by Western blotting method.Results The miR-21and mRNA expression levels in DU145cell line were significantly higher than those in PC-3and LNCaP cell lines(P<0.05).Up regulated miR-21expression could inhibit apoptosis of DU145cells,enhance migration ability of DU145cells and up regulate protein expression level of Wnt signaling pathway(P<0.05).Conclusion The miR-21can affect biological behavior of prostate cancer by Wnt signaling pathway.
