四爿藻培养模式的筛选及其培养基的优化 被引量:1

摘要 [目的]为了提高热带四爿藻的生长速率。[方法]以"宁波大学3#微藻培养基"为基础,分别添加有机碳(葡萄糖和乙酸钠)对四爿藻进行自养、兼养及异养培养,筛选出促进四爿藻快速生长的培养模式,并通过单因子及正交试验,优化其培养基配方。然后用优化培养基与"宁波大学3#微藻培养基"对比培养四爿藻。[结果]乙酸钠是四爿藻兼养培养的适宜有机碳,其最佳的乙酸钠浓度为6 g/L。获得了四爿藻的兼养培养基为:6 g/L CH3COONa、50 mg/L NH2CONH2-N、2 mg/L Ca(H2PO4)2-P、1 mg/L Fe SO4-Fe、1 mg/L Vitamin B1、0.000 5 mg/L Vitamin B12和0.5 mg/L Vitamin H。优化兼养培养基与"宁波大学3#微藻培养基"对比培养四爿藻的结果表明,培养第7天,兼养培养收获的藻细胞密度、干重分别达到3.84×106cells/m L及1.14 g/L,分别是"宁波大学3#微藻培养基"的2.83倍、3.12倍。[结论]四爿藻进行兼养培养可获得高密度培养。 [Objective]To improve the growth rate of Tetraselmis chui.[Method]Based on“Ningbo University#3medium”,organic carbon(C6H1206and CH3C00Na)was added for the autotrophic culture,mixed culture and heterotrophic culture,respectively,and the mixotrophy medium of T.chui was further optimized by single factor test and orthogonal test.And then,the results of mixotrophic and autotrophic cultures were compared.[Result]CH3C00Na was the better organic carbon applicable for the mixotrophic culture on T.chui.The optimization of mixotrophy medium was6g/L CH3C00N a,50mg/L NH2CONH2-N,2mg/L Ca(H2P04)2-P,1mg/L FeS04-Fe,1mg/L Vitamin Bj,0.0005mg/L Vitamin B12and0.5mg/L Vitamin H.The cell density and dry weight of the‘‘mixotrophy medium”was up to3.84x106cells/mL and1.14g/L after culturing seventh day,respectively,being2.83/3.12times higher than the cell density and dry weight of“Ningbo University#3”medium.[Conclusion]T.chui can be cultured with high cell density through mixotrophic culturing.
